High speed freeze centrifugeIt is the core equipment for separating samples in laboratories such as biology, medicine, and food. It achieves solid-liquid separation by centrifugal force generated by high-speed rotation, while protecting the activity of thermosensitive samples through freezing function. To ensure the safe and stable operation of the equipment and accurate experimental results, the following operating procedures must be strictly followed.
1、 Preparation before operation
Equipment inspection: Firstly, observe whether the body is stable and whether the power and data cables are firmly connected without damage; Turn on the power switch of the device, enter the control panel interface, confirm that the temperature display, speed display and other parameters are normal, and there are no error prompts; Check if the refrigeration system can cool properly. You can set the pre cooling temperature (such as 4 ℃) in advance and wait for the temperature to stabilize before proceeding with subsequent operations.
Sample and centrifuge tube preparation: Select centrifuge tubes that are compatible with the equipment rotor to ensure that there are no cracks, aging, or other issues with the centrifuge tubes; Measure the sample according to the experimental requirements, paying attention to the moderate volume of the sample inside the centrifuge tube (generally not exceeding 2/3 of the centrifuge tube volume), to avoid sample overflow and contamination of the rotor during centrifugation; If multiple samples are centrifuged simultaneously, the samples need to be balanced to ensure that the weight difference of each centrifuge tube does not exceed the equipment requirements (usually ≤ 0.1g). Balancing can be achieved by adding an equal amount of blank reagent.
Rotor selection and installation: Select the appropriate rotor based on the centrifugal requirements of the sample (speed, capacity), check for scratches, corrosion on the rotor surface, and foreign objects in the rotor holes; Grasp both sides of the rotor with both hands and smoothly place it into the centrifugal chamber of the device, ensuring that the bottom of the rotor is completely in contact with the shaft. Tighten the rotor fixing nut clockwise to avoid loosening and causing abnormal noise or danger during high-speed rotation.
2、 Core operational steps
Sample loading: Place the balanced centrifuge tube symmetrically into the rotor hole, gently press it to the bottom, and ensure it is placed in place; If only a portion of the centrifuge tubes are loaded, they should be placed according to the principle of "symmetrical distribution". For example, when only two 6-hole rotors are used, they should be placed in opposite hole positions to avoid imbalance during high-speed rotation of the rotor.
Parameter setting: Close the centrifuge chamber cover, tighten the cover buckle to prevent air leakage; Set parameters on the control panel in sequence: ① Temperature: Set according to the characteristics of the sample (such as 4 ℃ for biological samples and -20 ℃ for special samples), and set to "room temperature" if freezing is not required; ② Speed: Strictly set according to the experimental plan, and shall not exceed the maximum allowable speed of the selected rotor (rated speed marked on the rotor); ③ Time: Set the centrifugation time according to the separation requirements (such as 5 minutes, 10 minutes). If continuous centrifugation is required, the "continuous operation" mode can be selected; ④ Acceleration/deceleration level: For general samples, medium speed acceleration and deceleration are selected, while for sensitive samples that are prone to delamination, low-speed acceleration and deceleration can be chosen.
Start running:High speed freeze centrifugeAfter completing the parameter settings, click the "Start" button on the control panel, and the device will start pre cooling (if set to low temperature). When the temperature reaches the set value, it will automatically start accelerating rotation; During operation, closely observe the equipment's operating status. If there is severe vibration, abnormal noise, or error, immediately click the "emergency stop" button and wait for the equipment to completely stop before troubleshooting.
3、 Organize after operation
Equipment shutdown: After the centrifugation time is over, the equipment automatically decelerates to a stop. After the rotor is completely stationary (the control panel displays "0 rpm"), open the centrifuge chamber cover; Do not open the cover when the rotor is not completely stopped to avoid danger caused by rotor inertia.
Sample extraction and processing: Use both hands to smoothly remove the centrifuge tube to avoid tilting and mixing the sample; Observe the separation effect of the sample (such as the separation of clear liquid and precipitate), and collect the sample according to the experimental requirements; If there is sample spillage, promptly wipe and clean the rotor and centrifuge chamber with gauze dipped in 75% ethanol.
Equipment cleaning and maintenance: Remove the rotor, wipe the surface and holes of the rotor with a dry soft cloth to remove residual liquids and impurities, and then place the rotor in a dedicated storage rack; Wipe the inside of the centrifuge chamber with clean gauze to keep it dry and prevent the growth of mold; Turn off the device power, unplug the power cord, and tidy up the experimental table.
4、 Safety precautions
It is strictly prohibited to use damaged or aged centrifuge tubes to avoid bursting during centrifugation; After loading the sample into the centrifuge tube, the lid should be tightly closed to prevent leakage.
The rotor installation must be firm, and the speed should not exceed the rated speed of the rotor when setting parameters, otherwise it may cause rotor damage or equipment failure.
During equipment operation, the centrifugal chamber cover must not be opened arbitrarily, and rotating parts must not be touched; In case of abnormal situations, priority should be given to using the "emergency stop" function, and do not forcefully cut off the power.
Regularly clean the centrifugal chamber and rotor, and keep the equipment dry when not in use for a long time to avoid component corrosion caused by humid environments.
