Introduction
Cellulose sulfate is a biocompatible ester substance, and its microcapsules are prepared by dropping a polymer solution into a fixative containing synthetic poly cationic poly diallyldimethylammonium chloride (synthetic poly cationic poly DADMAC). Two polymers with opposite charges interact rapidly at the interface, and finally form a hydrogel film with strong mechanical stability by one-step method. Due to the strong interaction between sulfonic acid groups, these microcapsules have excellent mechanical properties and do not trigger immune reactions. In addition, the membrane structure is uniform, the pore size distribution is narrow, and the pore size can be pre-set.
This technology is mainly used for encapsulating animal cells and stem cells, applied in the fields of medicine and biotechnology, and has entered the clinical trial stage for cell transplantation in the human body to treat various diseases. It can also be used to encapsulate active pharmaceutical ingredients (APIs).
Research objective: To use a one-step process to produce cellulose sulfate polyDADMAC microcapsules, in order to distinguish them from other cell packaging processes that require multi-step operations, such as the alginate poly-L-lysine process

Experimental process and parameters
Remove the pre filter from the microcapsule granulator.
Dissolve 2g of cellulose * in 100mL of water. Dissolve it using a blender (e.gFigure 1). Let the solution settle until it becomes clear and internal bubbles are released. Bubbles can be accelerated for removal by placing them in an ultrasonic bath or vacuuming.
Add 8g of polyDADMAC (molecular weight 35 kDa) to 200mL of water and stir until dissolved.
The material to be embedded needs to be mixed with cellulose and added to the solution. During the capsule production process, these materials are wrapped in a cellulose core, which is then wrapped in a polyDADMAC film.
Take 20mL of cellulose * solution. After forming a stable series of droplets, start dripping the curing solution (polyDADMAC) for production (hardening), using electrostatic charge to disperse the droplets and prevent collisions. After the cellulose * droplet falls into the curing solution, let it harden for at least 30 minutes (starting point T=0: the moment when the last droplet falls into the curing solution). Wash the capsule with plenty of water to remove any unreacted polyDADMAC that may exist around the particles.

Experimental Conclusion
Buqi Microcapsule Granulator B-390 and B-395 ProCapable of producing spherical cellulose sulfate polyDADMAC microcapsules in a single step processThe particles produced haveExtremely narrow size distributionThese capsules can also be produced under sterile conditions using the reaction vessel of B-395 Pro, forEmbedding of animals and stem cells for biomedical applications.
Many studies have shown that,Compared to alginate poly L-lysine capsule systems, these types of capsule systems function better as they do not trigger immune responses and form more stable structures.
The microcapsule granulator can use nozzles of different sizes, and the size of cellulose sulfate capsules can be selected within the range of 400-2500 μ m. The capsule size can also be changed by using polyDADMAC with different molecular weights.
