The installation of Raman microscope components must strictly follow the standard procedures to ensure equipment performance, safety, and service life. The following provides a detailed description of pre installation preparation, installation of core components, optical path debugging, environmental control, and other aspects:
1、 Preparation work before installation
1. Environmental requirements
-Temperature and humidity: The installation site should maintain a constant temperature (recommended 25 ± 2 ° C), with a relative humidity controlled between 20% and 80%. Avoid direct sunlight, air conditioning vents, or near water sources to prevent equipment from being affected by moisture or temperature differences that may affect accuracy.
-Cleanliness: The operating area should be dust-free and vibration free, and the desktop should be flat and stable. It is recommended to equip it with anti-static pads and dust covers.
-Power inspection: Confirm that the input voltage is consistent with the equipment requirements (such as 220V AC/50Hz), and that the grounding is good (the voltage difference between the neutral and ground wires is less than 1.5V).
2. Tools and Protection
-Prepare specialized tools such as a screwdriver, tweezers, lens paper, dust-free cloth, lens cleaner, etc.
-Operators need to wear lab coats, gloves, and goggles to avoid oil or sweat from coming into contact with optical components.
2、 Installation steps of core components
1. Installation of eyepiece and objective lens
-Eyepiece: Remove the protective cover of the eyepiece tube, insert the eyepiece vertically and rotate it to a fixed position to ensure that the eyepiece is in contact with the tube. Attention should be paid to adaptability when replacing eyepieces with different magnifications.
-Objective lens: Rotate the objective lens converter in the order of low magnification to high magnification, hold the edge of the converter with your fingers and rotate it to avoid damaging the threads by directly pulling the objective lens. After installation, it is necessary to check whether the top of the converter spring matches the wedge-shaped groove and hear a "click" sound to confirm locking.
2. Lighting system and light path debugging
-Light bulb installation: After turning off the power, wear gloves to remove the halogen light bulb and avoid touching the glass surface. Adjust the filament position after installing the lamp holder:
-Use a telescope to observe the filament image, and adjust the horizontal ("-" hole) and vertical screws to evenly fill the objective aperture stop with the filament image.
-Install frosted glass sleeves to evenly scatter light and enhance field of view brightness.
-Focusing lens and filter: The aperture stop of the focusing lens needs to match the numerical aperture of the objective lens (such as 0.6 aperture for a 40 x objective lens). The filter should be selected according to the laser wavelength, and attention should be paid to the direction and angle during installation to avoid interference from reflected light.
3. Sample stage and clamping device
-Adjust the lifting knob of the stage to ensure that the distance between the objective lens and the stage is sufficient (about 5mm) to avoid collision.
-Low temperature or strong magnetic field experiments require the pre installation of refrigeration chips or magnetic control modules, and the calibration of displacement parameters through software.
3、 Optical path calibration and testing
1. Kohler lighting adjustment
-Select a 10 × objective lens, remove the eyepiece and insert it into the centering telescope, adjust the position of the lamp chamber to make the filament image clear.
-Switch to a 40 × objective lens and fine tune the position of the filament to fill the field stop.
-After restoring the eyepiece, focus the sample to clarity and observe the uniformity of the light spot.
2. Laser path collimation
-Turn on the laser light source and adjust the reflector to make the beam pass through the beam expander and filter before entering the objective lens vertically.
-Use infrared cards or fluorescent samples to verify the position of the light spot, ensuring that the laser center coincides with the optical axis of the objective lens.
4、 Safety and maintenance precautions
1. Electrical safety
-Before replacing the light bulb, fuse, or plugging in the power supply, the power must be cut off and live operation is strictly prohibited.
-Use the original factory specified power cord to avoid overload or voltage fluctuations.
2. Maintenance of optical components
-Lens cleaning requires blowing a ball to remove dust, using a cotton swab dipped in alcohol for one-way wiping, and using organic solvents such as xylene is prohibited.
-Immediately wipe the oil containing objective lens after each use to prevent the oil from solidifying.
3. Long term storage management
-Unplug the power cord, cover with a dust cover, and regularly power on for dehumidification (once a month).
-Record device status and archive maintenance logs.
