As a precision analytical instrument, the stability and service life of Raman microscopy greatly depend on scientific maintenance methods. The following is a detailed description of the maintenance method for Raman microscopy:
1、 Daily maintenance
1. Cleaning and dust prevention
-Lens cleaning: After each use, gently wipe the surface of the objective lens, eyepiece, and reflector with dust-free paper or specialized optical lens paper to avoid dust accumulation that affects the optical path. For stubborn stains, a small amount of anhydrous ethanol can be used for cleaning, but it is necessary to avoid liquid infiltration into the inside of the lens.
-Sample table cleaning: Clean the residual powder or liquid on the sample table in a timely manner to prevent contaminants from corroding the table or interfering with subsequent testing.
-Dust cover application: When the instrument is not in use, it needs to be covered with a dust cover to prevent dust from entering the optical system in the laboratory environment.
2. Optical path inspection and calibration
-Daily pre startup check: Turn on the white light switch and observe whether the laser is accurately incident on the sample position. If there is any deviation, adjust the optical path.
-Regular calibration: Focus and optical path calibration are performed weekly using standard samples (such as monocrystalline silicon wafers). By adjusting the coarse and fine adjustment knobs, the surface of the silicon wafer can be imaged clearly, and the objective lens can be switched to a high magnification lens (such as 50x) to further optimize the focus.
3. Software and hardware shutdown process
-Proper shutdown: After the test is completed, the laser power should be turned off first. After the laser cools down, the main power should be turned off to avoid instantaneous power failure and damage to components.
-Software exit: Before closing the data collection software, it is necessary to save the experimental data and release system resources to prevent software abnormalities from causing system crashes.
2、 Periodic maintenance
1. Replacement of key components
-Filter and slit cleaning: Check the filter and slit once a month, use compressed air to remove surface dust, and if necessary, use non-woven fabric dipped in alcohol to gently wipe.
-Laser status monitoring: Detect laser power fluctuations every six months. If the output power is lower than 80% of the initial value, contact the manufacturer for replacement or repair.
2. Mechanical structure lubrication
-Sample stage transmission system: Add a small amount of precision lubricating oil to the moving track of the sample stage every quarter to ensure smooth sample movement and accurate positioning.
-Flexibility inspection of focusing knob: Regularly check the gear engagement of the coarse/fine adjustment knob to avoid jamming and affecting the focusing accuracy.
3、 Environmental control requirements
1. Temperature and humidity management:
-The laboratory temperature should be controlled at 20-25 ℃, and the humidity should be maintained at 40% -60%. High temperature can easily cause deformation of optical components, while high humidity may trigger mold growth or corrosion of metal parts.
-Avoid placing the device near direct sunlight or air conditioning vents to reduce the impact of temperature fluctuations on calibration.
2. Shock absorption and anti magnetic:
-The desktop where instruments are placed should be equipped with shock-absorbing pads and kept away from large mechanical equipment or magnetic sources (such as magnets and electromagnetic coils) to prevent vibration or magnetic field interference with optical path stability.
4、 Operating standards and precautions
1. Sample processing specifications
-Solid state samples: Ensure a smooth surface and avoid scratching the objective lens with sharp edges. Double sided tape can be used to fix the sample, but the amount of tape should be controlled to prevent residual contamination.
-Liquid sample: It should be sealed in a quartz colorimetric dish or a dedicated liquid pool to prevent solvent evaporation or leakage from corroding the instrument
2. Test parameter optimization
-Select laser power and acquisition time based on sample characteristics. Excessive power may burn the sample, while insufficient power may result in weak signals; Accumulating too many times can introduce thermal noise.
-When focusing, it is necessary to avoid prolonged laser irradiation on the same area to prevent sample decomposition or carbonization.
3. Handling of abnormal situations
-Signal attenuation: Firstly, check whether the sample is offset or defocused. If the optical path is normal, calibrate the spectrometer or check the laser status.
-Spectral distortion: It may be caused by environmental vibrations or sudden temperature changes, and testing should be stopped immediately to investigate environmental interference.
5、 Long term disuse maintenance
1. Sealing preparation:
-After turning off all power sources, blow the inside of the instrument with dry nitrogen and place a desiccant inside the box to prevent moisture retention.
-Record the equipment status before sealing (such as laser usage duration, filter cleanliness) for the purpose of verifying performance after restarting.
2. Restart and restore:
-After long-term shutdown, it is necessary to preheat for at least 30 minutes and recalibrate the optical path and spectrometer when starting up.
