Fully automatic and ultra sensitive single-cell multifunctional protein analysis systemThrough IsoCode chip detection technology, complete biomolecular and functional information of thousands of individual living cells can be captured simultaneously, and the cells are in a survival state throughout the analysis process, allowing real-time monitoring of the expression of up to 32 proteins secreted by live cells at the single-cell level.
The working principle of the IsoLexis system is mainly based on its unique microfluidic chip technology and automated detection process, which can achieve high-throughput and multiplexed proteomic analysis at the single-cell level. The following is its core workflow:
Single cell capture and culture:
The system uses microfluidic chips (IsoCode chips) to separate individual cells into microchannels, and achieves uniform cell distribution and culture environment through fluid dynamics design. This step avoids the problem of requiring multiple manual operations and cleaning stations in traditional methods.
Protein secretion and detection:
After cells secrete proteins in the microchamber, the system adds ELISA reagents through an automated process and uses a sandwich method to perform high-sensitivity detection of secreted proteins (such as cytokines). Each microcavity can analyze up to 32 proteins, with a detection range covering 2-2000 pg/ml.
Imaging and Data Analysis:
The system is equipped with advanced optical components to image cell and protein signals within the microcavity, and directly import the data into the IsoPeak software platform. This software quantifies protein expression at the single-cell level through algorithms and generates functional protein profiles (such as the Cell Multifunctional Index PSI) for identifying cell subpopulations and functional states.
