preparation:
Confirm component integrity: Ensure that the chromatography column, adapter, and all related accessories (such as O-rings, sieves, pipelines, etc.) are complete and undamaged.
Cleaning components: Use appropriate cleaning agents (such as 20% ethanol) to clean the chromatography column, adapter, and accessories, ensuring that there is no dust, dirt, or residue.
Check compatibility: Confirm that the interface size of the adapter matches the interface size of the chromatography column.
Installation steps:
Fix the chromatography column vertically on a fixture or iron frame.
If the chromatography column has a lower column head, install the lower column head first and tighten the sealing ring tightly.
Add an appropriate amount of column loading solution (such as distilled water, 20% ethanol, etc.), with a liquid level height of generally 1-2cm.
Connect the adapter pipeline to the syringe and remove any air bubbles from the adapter pipeline.
Place the adapter vertically on the chromatography column and tighten the fixing cap clockwise to ensure a secure connection.
Adjust the height of the adapter according to the experimental requirements.
Connection and debugging:
Connect the column head tubing to the chromatography system (such as AKTA).
Operate the equipment according to the recommended flow rate in the packing manual and perform column pressing operation.
After the compression column is completed, perform column efficiency inspection to ensure that the column efficiency meets the experimental requirements.
