1、 Preparation work

Cleaning components: Use appropriate cleaning agents (such as 20% ethanol) to clean the chromatography column, adapter, and accessories, ensuring that there is no dust, dirt, or residue.
Check compatibility: Confirm that the interface size of the adapter matches the interface size of the chromatography column to ensure a tight connection.
2、 Installation steps
Fixed chromatography column: Fix the chromatography column vertically on a fixture or iron platform, ensuring that the column tube is not tilted and perpendicular to the ground.
Install lower column head: If the chromatography column has a lower column head, first install the lower column head and tighten the sealing ring. Ensure that the bottom column head sieve plate is moist and expel any air bubbles.
Add column loading solution: Add an appropriate amount of column loading solution (such as distilled water, 20% ethanol, or 150mM NaCl solution) to the chromatography column, with a liquid level height of generally 1-2cm.
Install adapter:
Connect the adapter pipeline to the syringe and remove any air bubbles from the adapter pipeline.
Rotate the adjustment cap counterclockwise to rotate the fixed cap and adjustment cap to the bottom of the adjustment rod.
Open the bottom pipeline plug, place the adapter vertically on the glass tube, and tighten the fixing cap clockwise. Ensure that the entire operation is completed in a straight line to avoid introducing bubbles.
Adjust the height of the adapter: According to the experimental needs, adjust the height of the adapter to maintain an appropriate distance from the gel bed interface of the chromatography column.
3、 Compression column and debugging
Connect the chromatography system: Connect the column head tubing to the chromatography system (such as AKTA, etc.), ensuring a secure connection and no bubbles.
Column pressing: Operate the equipment according to the recommended flow rate in the packing manual and perform column pressing operations. During the compression process, pay attention to the changes in the interface of the glue bed and adjust the height of the adapter in a timely manner.
Debugging and inspection: After the compression column is completed, perform column efficiency inspection. By measuring the spectrum, we can understand the loading situation of the chromatography column and ensure that the column efficiency meets the experimental requirements.
4、 Precautions
Avoid overpressure: During the process of column installation and compression, if the optimal pressure column flow rate conflicts with the pre column pressure, the pressure should prevail to avoid overpressure causing damage to the adapter or chromatography column.
Prevent bubbles: During the entire installation process, special attention should be paid to preventing bubbles from entering the chromatography column or adapter. Once bubbles are found, the operation should be stopped immediately and the column should be reinstalled.
Keep clean: During and after installation, keep the chromatography column and adapter clean to avoid dust and dirt affecting the experimental results.
