1 Wet state antibacterial test device YY/T 0471.5 glass instrumentscope
This section of YY/T0471 specifies the test for evaluating the antibacterial properties of contact wound dressings claiming to have antibacterial properties
Method 1).
This test method involves microbial detection operations and should be conducted by trained personnel in a biosafety laboratory.
2 Terms and definitions
The following terms and definitions apply to this document.
2.1
Dry state low moisture condition
The state where both sides of the dressing are not wet. The dressing in this state has no exudate or minimal exudate from the wound, while the outer surface of the dressing is in
Dry state.
Note: If the dressing does not claim to be water-resistant, it is considered that the outer surface is in a dry state.
2.2
Semi wet condition
One side of the dressing is in a wet state. The dressing in this state has exudate from the wound, while the outer surface of the dressing is in a dry state, that is, "external"
Dry and wet inside "; Alternatively, the wound may have no exudate or minimal exudate, while the outer surface of the dressing is expected to be in a moist state, known as' wet on the outside and dry on the inside '.
Note: If the dressing does not claim to be water-resistant, it is considered that the outer surface is in a dry state; The dressing claims to be water-resistant and is considered to have a moist outer surface.
2.3
Wet condition
Both sides of the dressing are wet. The dressing in this state has exudate from the wound, and the outer surface of the dressing is expected to be moist
state
Note: If the dressing claims to be water-resistant, it is considered that the outer surface is in a moist state.
3 Reagents and materials
3.1 Serratia marcescens 2);
3.2 Nutrient broth culture medium;
3.3 Nutrient agar medium;
3.4 Sterile physiological saline solution;
3.5 Sterile swabs.
1) The antibacterial components in the dressing may have an impact on the test results in this section. Appendix A provides application instructions for this section.
2) The standard strains of strains recognized by domestic and foreign bacterial preservation institutions, such as ATCC8100, CICC10187, etc. Some sticky sand bacteria can appear on the surface of agar
The appearance of red colonies, such as CICC10187, can be used to check for the growth of Serratia marcescens. This information is provided for the convenience of our
Some users.

4.1 Challenge vaccine preparation
Pick the colony of Serratia marcescens from the working strain of Serratia marcescens on a sloping surface, streak and inoculate it on a nutrient agar plate at 26 ℃
The challenge inoculum was prepared by overnight cultivation under conditions, and used for dry and semi wet (external dry and internal wet) antimicrobial assays. Prepared tablet
Suitable for daily use.
4.2 Preparation of challenge bacterial solution
Select single colonies of Serratia marcescens from the challenge inoculum plate prepared in section 4.1 and inoculate them into nutrient broth medium. Incubate at 26 ℃
Night cultivation was conducted to obtain a challenge bacterial solution with a concentration of approximately 109 CFU/mL, which was used for semi wet (external wet dry internal) and wet state antibacterial tests. Ready to prepare
The challenge bacterial solution should be used on the same day.
5 Dry state antibacterial properties
5.1 Experimental apparatus
Figure 1 is a schematic diagram of the dry state antibacterial test device. The challenge room is used to hold challenge inoculants, while the sampling room is used for wiping and sampling. Before the experiment,
Use confirmed sterilization procedures to sterilize the experimental device for backup.
