Column selection for SEC analysis of lipid nanoparticles (LNP)

Summary of the plan:

In 2018, the world's first nucleic acid drug using lipid nanoparticles (LNP) to deliver siRNA was approved. Recently, due to the development of new lipid components and production methods, mRNA vaccines targeting SARS-CoV-2 coronavirus have been developed for LNP delivery. Recently, in a research paper titled "Enabling online determination of the size dependent RNA content of lipid nanoparticle based RNA formulations" published in the J. Chromatogr. B journal, the authors and their team believe that the size and mRNA content (payload) of LNP are crucial for understanding drug efficacy and side effects. Therefore, size exclusion chromatography TSKgel G6000PWXL-CP was used, and dynamic light scattering (DLS) detectors were directly connected to UV detectors, RI detectors, and MALS detectors for online particle size analysis of LNP.
