Passage operation process

1. Try to clean the T25 bottle of original culture medium as much as possible;
2. Add 3-4ml of room temperature PBS and gently rinse the cells for 10-20 seconds, then remove the washed PBS;
Add about 1ml of pancreatic M to T25 bottle, gently shake the culture bottle to spread the pancreatic M evenly on the bottom cell layer of the bottle;
4. Put the culture bottle into a 37 degree incubator for digestion;
5. Stop pancreatic M digestion by adding 2-3 ml of culture medium until the intercellular space becomes larger but does not detach;
6. Mix the cells well, centrifuge at 900rpm for 3-5 minutes, and discard the supernatant;
7. Gently resuspend the cells in new culture medium and evenly distribute them into new culture bottles;
8. Supplement the culture medium and place the culture bottle back in the incubator for static cultivation;

Precautions

There is a significant difference in the digestion time of pancreatic M among different brands, so it is important to strictly control the digestion time;
When digesting passaged cells, it is important to blow them gently and avoid producing too many bubbles to avoid affecting the cell state.
