Alkaline phosphatase can be coupled to the free amino groups of antibodies or proteins using glutaraldehyde as a crosslinking agent.

Different samples have different optimal molar ratios. For antibodies, the molar ratio of the labeled sample to AP should not exceed 1:2. Other samples can be tried in different proportions.

Customization - By changing the molar ratio, reaction buffer, pH value, and other parameters, different levels of AP coupling can be achieved.

Precautions during use:

Please note that components containing amino groups are not allowed in the buffer, as this can affect the coupling effect.

If it is necessary to dilute the conjugate, avoid using phosphate rich buffer solutions such as PBS, and use phosphate free buffer solutions such as TBS.

If long-term storage is required, please pack and store in the dark at -20 ° C to avoid repeated freezing and thawing. The same volume of glycerol can be added.

Background: Alkaline phosphatase(ALP or AKP is an enzyme that can dephosphorylate corresponding substrates by hydrolyzing phosphate monoesters to remove phosphate groups from substrate molecules and generate phosphate ions and free hydroxyl groups. These substrates include nucleic acids, proteins, alkaloids, etc. The process of removing phosphate groups is called dephosphorylation or dephosphorylation. Alkaline phosphatase is a type of phosphatase, but it is not a single enzyme, but a group of isoenzymes. Alkaline phosphatase can bind to molecules to be labeled, and when incubated with appropriate substrates, it generates labeled molecule conjugates with colored, fluorescent, or luminescent derivatives that we can detect and quantify. AP is commonly used in conjugates to determine the presence of molecular targets, and is commonly used in experiments such as ELISA, protein immunoblotting, and immunohistochemistry. Due to the competitive inhibition of alkaline phosphatase activity by higher concentrations of inorganic phosphorus, phosphate rich systems such as PBS should not be used in immune experiments, otherwise it will cause a higher background. Therefore, phosphate free systems such as TBS can be used.
