Product Overview:

人水蛭素(HRD)Specially designed for monitoring anticoagulant therapy, accurately detecting levels of hirudin in serum. Hirudin is a potent enzyme inhibitor widely used in anticoagulant therapy for thrombotic diseases, adapted to serum samples, and provides data support for clinical dosage adjustment and bleeding risk assessment.

Kit composition:

Pre coated microplate: 96 well or 48 well specification, coated with RCA capture antibodies.

RCA standard: Provides original standard samples for drawing standard curves.

Label detection antibody: a detection antibody that specifically recognizes RCA.

Streptomycin HRP conjugate: used for signal amplification.

Substrate solution (TMB): Chromogenic substrate, turns blue when exposed to HRP enzyme.

Termination solution: Terminate the reaction, and the color of the solution changes from blue to yellow.

Washing buffer (10 x concentrated solution): used for cleaning microplates.

Sample: Used for diluting samples and standards.

Sealing film: prevents cross contamination.

Sample requirements:

Sample type:Serum, plasma, tissue homogenate, cell culture supernatant, etc.

Sample processing:

Serum/plasma:After collection, let it settle and centrifuge to remove sediment. Pack and store at -80 ℃ to avoid repeated freezing and thawing.

Tissue homogenateAdd the lysate homogenate in proportion and centrifuge to obtain the supernatant.

Cell culture supernatant: Centrifuge to remove debris and detect.

Attention:Avoid hemolysis or high-fat samples, fresh samples are preferred, and long-term storage requires -80 ℃.

Operation steps:

1、 Prepare reagents:

Equilibrate the reagent kit to room temperature 20 minutes in advance, and dilute the concentrated washing solution with distilled water.

2、 Sample addition:

Gradient dilution of standard samples: Dilute the original standard sample proportionally.

Sample processing: Take 50 μ L directly or diluted and add it to the micropores.

3、 Nurturing:

Add samples/standards and incubate at 37 ℃ for 1 hour.

After washing 3 times, add labeled antibodies and incubate at 37 ℃ for 1 hour.

Wash again, add streptavidin HRP, and incubate at 37 ℃ for 30 minutes.

4、 Color rendering and termination:

Add TMB substrate and incubate in the dark for 10-30 minutes.

Add termination solution to stop the reaction.

5、 Testing:

Immediately measure the absorbance (OD value) using an enzyme-linked immunosorbent assay (ELISA) reader at a wavelength of 450nm.
