In vitro activity:

Method: Used for in vitro bioluminescence analysis Preparation of D-Luciferin potassium:

1. Prepare 200 × D-Luciferin stock solution (30 mg/mL) with sterile water, gently mix until completely dissolved. After preparation, it can be used immediately or stored separately at -20 ℃.

2. Prepare 1 × D-Luciferin (150 µ g/mL) working solution in preheated tissue culture medium.

3. Suck out the culture medium for cultivating cells.

4. Add 1 × D-Luciferin solution to the cells before imaging. Cultivating cells at 37 ℃ for a short period of time before imaging can increase signal.

Internal activity:

Method: Used for in vivo bioluminescence analysis Preparation of D-Luciferin potassium:

1. Prepare fresh 15 mg/mL D-Luciferin stock solution using DPBS. Filter and disinfect with a 0.20 μ m filter.

2. Inject 10 µ L/g according to the weight of the mouse. Each mouse should be injected with 150 mg of D-Luciferin/kg body weight. For example, for mice weighing 10 g, injecting 100 µ L provides 1.5 mg of D-Luciferin.

3. Inject D-Luciferin intraperitoneally for 10-15 minutes during imaging. Fluorescein kinetics studies should be conducted for each animal model to determine peak signal time.
