SID4 Bio is a quantitative phase detection instrument based on four wave shear interferometry technology launched by Phase. The equipment can be directly used with a general microscope to obtain the quantitative phase distribution of biological cells.

In biomedicine, intracellular fluid and tissue fluid are usually transparent, making it difficult to distinguish solely based on color differences or grayscale in images, and even unable to obtain contrast images due to transparency; So it is usually necessary to fluorescently label cells in order to detect and sort them; However, cell labeling or staining requires pre-treatment, which can interfere with cell activity and growth, and even lead to cell death, making it impossible to detect live cells; In order to excite fluorescence, an external excitation light source is usually required. To improve excitation efficiency, focused laser is usually used for excitation, which can further stimulate cells and some may even cause optical toxicity; Quantitative phase detection technology utilizes the difference in refractive index between cell fluid and surrounding tissue fluid to achieve label free imaging, without the need for sample pretreatment, achieving plug and play functionality and minimal intervention in cell activity, allowing for real-time recording and detection of cell activity; Moreover, as a phase testing technique for self chromatic aberration, it is possible to select wavelengths with low optical toxicity for illumination or broad-spectrum illumination based on the sample, in order to minimize external interference on cells as much as possible.
