COSMOSIL 5C18-PAQ chromatographic column

1. Method for removing buffer, salt, and acid from the chromatographic column: Rinse with a mobile phase free of buffer, salt, and acid for 10-15 minutes.
   Example: When the mobile phase is methanol/phosphate buffer (20mmol/L)=50/50, rinse with methanol/water=50/50

2. The method of flushing the attached substances in the chromatographic column and the solution when the baseline is unstable

• When the sample is not protein, rinse with methanol or *.

• When the sample is protein, use 0.1%C2HF3O2Mix 50-70% acetonitrile/waterwash

1. Short term (several days) storage:
   Rinse with a mobile phase without buffer, salt, or acid for 10-15 minutes. preservation.
   Example: When the mobile phase is methanol/phosphate buffer (20mmol/L)=50/50, rinse with methanol/water=50/50

2. Long term (over 1 month) storage:
   After use, clean the chromatography column with a mobile phase free of acid and salt, then replace the mobile phase with acetonitrile/water=70/30 or methanol/water=70/30, and store.

It can be used except for alkaline solutions and strong acid solutions (pH below 1.5).(Strong alkaline solution will dissolve silica gel, strong acid solution will cause the stationary phase to detach)
Points to note:
Excessive solvent viscosity can cause pressure rise, please control the pressure below 20Mpa.After using acidic mobile phase or high freezing point solvents, the chromatography column must be cleaned.

• In general, a buffer concentration of 0.005~0.02 mol/L is sufficient.

• The mobile phase must be filtered through a membrane below 0.5 μ m before use.

• When analyzing proteins in reverse phase mode, please use a large pore size chromatography column (pore diameter 300 Å)COSMOSIL Protein - R（粒径5μm）。
