Product Description
HL-SAN nuclease is a high salt activity nuclease that can efficiently degrade single stranded and double stranded DNA/RNA under conditions of up to 1 M NaCl. Its unique salt tolerance and efficient degradation ability make it an ideal tool for biopharmaceuticals, vaccine production, and molecular biology research.
Key Features
*High salt activity: Maintain high activity under 0.2 M -1 M NaCl conditions.
*Wide pH adaptability: It exhibits activity within the pH range of 5.0-9.0.
*Efficient degradation: It can rapidly degrade DNA/RNA to oligonucleotides of ≤ 5 bp.
*Low protein residue: Suitable for GMP production environment and meets high purity requirements.
Instructions for Use
1. Resolution and dilution
*Before use, place the freeze-dried powder on ice to balance for 10 minutes.
*Dissolve in sterile deionized water or buffer solution (such as Tris HCl, pH 7.5), with a recommended final concentration of 1 U/μ L.
*Further dilute according to experimental requirements (recommended working concentration range: 0.1-10 U/mL).
2. Experimental application
*DNA/RNA removal:
-Add 1-10 U/mL HL-SAN nuclease under 0.2 M -1 M NaCl conditions and incubate at 37 ° C for 30 minutes.
-Terminate the reaction by heating (65 ° C, 10 minutes) or EDTA.
*Vaccines and Cell Therapy:
-Used to remove residual host cell DNA/RNA during the production process, ensuring product safety.
3. Precautions
*Avoid contamination: After reconstitution, package and store separately to avoid repeated freeze-thaw cycles leading to decreased activity.
*Reaction termination: Use EDTA (final concentration 5 mM) or heat (65 ° C, 10 minutes) to terminate the enzyme reaction.
*Waste disposal: Experimental waste must undergo high-pressure sterilization at 121 ° C for 30 minutes before being discarded.
quality control
*Activity detection: Enzyme activity is determined by fluorescence substrate method (1 U is defined as the amount of enzyme required to degrade 1 μ g DNA within 1 minute at 37 ° C).
*Purity testing: SDS-PAGE analysis, purity ≥ 95%.
*Batch consistency: COA (Certificate of Quality Analysis) is provided for each batch of products.
Safety Information
*Danger sign: No special danger (GHS standard).
*Emergency response:
-Skin contact: Immediately rinse with plenty of water.
-Inhalation/ingestion: Seek medical attention immediately and provide product information (CAS number: N/A).
Validity period and transportation
*Validity period: 2 years from the date of production (see label for details).
*Transportation conditions: Dry ice transportation to ensure low temperature throughout the entire process.
