The detection method based on microbial culture has been leading the monitoring of oilfield microorganisms for many years. The current industry standards and internal standards of various oilfield enterprises mostly evaluate the bactericidal effect of oilfield produced water fungicides based on the extinction dilution method (for example, Shengli Oilfield mainly refers to the technical requirements for fungicides used in oilfield produced water treatment in SY/T 5757-2010). But other tools such as the second-generation ATP luminescence methodATP oilfield microbial activity detection reagent kitAdenosine triphosphate (ATP) and qPCR (quantitative polymerase chain reaction) are becoming increasingly common in field and laboratory evaluations, and these tools can effectively compensate for the long detection cycle (7 days) of culture methods and the underestimation of actual microbial populations (ignoring 99.9% of other microorganisms besides specific microorganisms).
The correlation between second-generation ATP luminescence method and plate method
The experiment shown in the figure below selected oil containing samples (red) and non oil containing samples (blue) with different microbial concentrations for testing by plate method and ATP method, and then plotted the results of the two methods to verify their linear correlation and calculate the R2 value. From the graph, we can see that the R2 values between ATP and LB culture plate methods are both greater than 0.9, indicating a strong correlation between the two analysis methods.
It is worth emphasizing that the second-generation ATP luminescence method(ATP Oilfield Microbial Activity Detection Kit)Compared with the first generation ATP method, there are three significant improvements:
· Increase the analysis volume of the sample to 20mL to improve the stability and detection limit (LLD) of the analysis
· Added preprocessing steps to wash away organic matter and TDS from the sample to avoid interference
· Enzyme activity calibration was performed before each analysis to prevent loss of enzyme activity from affecting the results

Comparison of results between second-generation ATP luminescence method, extinction dilution method, and qPCR
The following figure shows the results of three different methods for analyzing microorganisms in actual water samples extracted from oil fields. The samples analyzed six times were all from the same sampling point, with a sampling interval of 1-2 months and a total time of 1 year. The result of qPCR method is the sum of the total number of bacteria and archaea; The extinction dilution method is the sum of SRB, APB, and GHB readings; ATP luminescence method refers to the total number of microorganisms. The results indicate that the extinction dilution method may greatly mislead the detection results of microorganisms (such as sample 5), while the results obtained by ATP luminescence method and qPCR method have strong consistency.

The following table shows the actual production data of Barnett Shale mining in the United States, and microbial analysis was conducted on samples from the wellheads of 8 different production wells. The ATP luminescence method and the unique dilution method were both completed on-site, while qPCR was used to analyze microbial samples trapped on the filter membrane in the laboratory. The results were expressed as the number of microorganisms per milliliter of water sample, with white indicating normal, yellow indicating high, and red indicating high. We can see that the results of ATP and qPCR methods are basically consistent, while the extinction dilution method may overestimate (production well 1) or underestimate (production well 3) the estimated microbial population.

summary
The second-generation ATP luminescence method in oilfield produced water(ATP oilfield microbial activity detection reagent kit)The microbial analysis results of qPCR method not only have good consistency with the results of extinction dilution method, but also can compensate for the long detection time and insufficient estimation of actual microbial population of extinction dilution method, which effectively compensates for the shortcomings of extinction dilution method. All three methods have their own characteristics, and the combination of multiple methods can help oil fields better manage microbial threats.
