Product Background:

cytokineIFNGamma can protect cells from viral infections and belongs to the interferon family. Numerous studies have shown that,IFNGamma secretion by cell types triggered by antigens, includingTCells, immaturityCD4+ TCells, macrophages, dendritic cells, andBcellIFN gammaIt plays an important role in triggering macrophages to fight against multiple microbial targets and is a multi effector molecule related to anti proliferation, pro apoptosis, and anti-tumor mechanisms. Due to the fact that this effector cytokine is considered the main effector factor of immunity, it has been used to treat various diseases.

Simpler: No need for antigen antibody reactions, the detection method based on small molecule chemical reactions is simple and efficient, and the reaction only takes a few minutes.

More sensitive: No need for antibodies, the detection dye is only 1/500 of BrdU antibody, which is easy to spread and can accurately detect even single proliferating cells.

Faster: No need to stay overnight, saving the complex and tedious steps of antigen antibody reactions, completing the entire testing cycle in just 2.5 hours.

More accurate: No DNA denaturation (acid hydrolysis, thermal decomposition, enzymatic hydrolysis, etc.) is required, which can effectively avoid sample damage caused by denaturation and ensure clear and intact cell nucleus edges.

More compatible: almost no damage to the sample, easier to label with multiple antibodies or fluorescent proteins simultaneously, able to simultaneously detect other characteristics of cells

Experimental procedure:

1. Cell culture

(Using a 6-well plate as a reference, other culture wells can be adjusted according to experimental needs.) Inoculate 1x10 ^ 5-3x10 ^ 6 cells per well into the 6-well plate and culture until normal growth stage.

Note: The amount of cell inoculation can be adjusted according to cell size, growth rate, and experimental treatment period.

2. Cell intervention or treatment

(Optional) Cells can be treated with drugs or other stimuli according to experimental needs.

3. Cell co incubation culture

According to the preliminary experimental results, dilute the solution with cell culture medium in proportion, or add an appropriate amount of solution to the well plate culture medium in proportion to co incubate with cells. At the same time, set the non addition group as the negative control group. Add diluted culture medium to each well and incubate for 2 hours to remove the old culture medium.

Notes:

Due to the use of a 96 well plate for testing, if your cells need to be cultured for 48 hours or longer, it is recommended not to skimp on the surrounding wells of the 96 well plate. These 32 wells cannot be used, and it is recommended to add sterile PBS to saturate the middle 64 wells with water. Because during the cell culture process, the water in the side holes evaporates quickly, causing concentration of the culture medium and drugs inside, which complicates the condition of the cells.

2. The solvent will solidify at low temperatures. Before use, please leave it at room temperature or incubate it in a water bath at 20-25 ℃ for a moment until it is completely melted before use.

3. Generally, it is effective to store at 4 ° C in the dark for two weeks. It should be stored at -20 ° C for a long time to avoid repeated freezing and thawing. Small doses should be divided and wrapped in light proof bags or black paper or tin foil to prevent decomposition. Especially when it turns gray green, it cannot be used again.

When the Formazan solution freezes or precipitates, it can be incubated in a 37 ℃ water bath to promote dissolution, and must be used after complete dissolution and mixing.

During incubation, it is necessary to avoid light.

6. It is carcinogenic, so be careful when using it and wear transparent film gloves if possible. The preparation needs to be sterile and sensitive to bacteria.
