Application background:

The function of FLEXCELL adjustable substrate stiffness culture consumables is to prevent hardness impact, which means that when cells transfer from a soft substrate to the surface of a regular polystyrene culture plate, stress reactions will occur due to changes in substrate hardness. The impact of cell hardness in this matrix hardness change is similar to the carbon source transfer experienced by bacteria when transferring from glucose to galactosecanCausing bacteria to increase the enzymes required for absorbing and metabolizing new carbon sources. The surface hardness of ordinary polystyrene culture plates is as high as 1 GPa, while the substrate hardness for cell growth is less than 1 GPa. The difference in hardness between the two is significant, and the stress response of cells caused by this cannot be ignored.

reference material:

The mechanical response to matrix hardness, shear stress, mechanical strain, cell morphology, and substrate topology during cell culture affects cell survival, proliferation, and differentiation (Kureel et al., 2019; Anderson et al., 2016; Engler et al., 2006; Gilbert et al., 2010; Lutolf et al., 2009; Murphy et al., 2014; Winer et al., 2009; Yeung et al., 2005). This is a challenge for clinical applications and cell-based therapies as they require prolonged expansion and large quantities of stem cells.

Data shows that various types of stem cells grown on soft 2D substrates (<1 kPa to 5 kPa) maintain their phenotype for longer periods of time than stem cells grown on hard polystyrene substrates (Disher et al., 2005; Engler et al., 2006; Buxbaum et al., 2010; Vertelov et al., 2016; Gerardo et al., 2019; Pagliari et al., 2021)

Cells grown on soft substrates affect cell morphology, gene expression, and stem cell differentiation. The therapeutic application of stem cells requires controlling their differentiation. Using CellSoft ® Soft matrix culture plates allow researchers to match the hardness of living surfaces with natural tissues.

CellSoft ® It is a special gel coating added to the surface of elastomer film or polystyrene. CellSoft ® Compared to uncoated glass or polystyrene culture plates, the substrate is softer and more ductile.

Product features:  Stiffness range: 1, 5, 10, 20, 40, 80 kPa

100mm culture dish, 6-well polystyrene bottom plate, BioFlex ® Stretchable 6-hole plate

 CellSoft ® BioFlex ® 6-well flexible culture plate for stretching cells on soft substrate

Pre disinfection, direct inoculation

Type I collagen matrix coating or untreated surface can be selected

Advantages of soft matrix for in vitro cell culture:

Match the in vitro matrix with natural hardness

Reduce the impact of substrate hardness on cells

• Stem cells have a longer retention time
