Nitrogen sealed liquid nitrogen tank: a sample safety fortress that locks "cryogenic" into the box
Date: 2025-12-14Read: 1

In cell therapy centers, stem cell banks, umbilical cord blood banks, and antibody drug development platforms, precious biological samples need to "sleep" in deep low-temperature environments below -190 ℃ for ten or even decades. Traditional liquid nitrogen tanks immerse samples directly in liquid nitrogen, although the temperature is extremely low, it is accompanied by risks such as cross contamination, tube bursting, and liquid nitrogen splashing. The emergence of the Dry Vapor Nitrogen Storage System is based on the concept of "liquid nitrogen does not immerse the sample, and the sample is in a dry area". This article will comprehensively interpret this "sample security fortress" from five dimensions: structural principles, core advantages, typical applications, selection points, and operation and maintenance experience.

1、 The technological transition from "wet" to "dry"

1. Liquid nitrogen may seep into the interior of the cryovial, causing tube bursting and cross contamination;

2. Operators need to come into contact with liquid nitrogen, which can easily cause frostbite;

3. After opening the lid, the liquid nitrogen evaporates violently, causing an instant increase in temperature and repeated freezing and thawing of the sample.

The nitrogen isolation type liquid nitrogen tank adopts a "liquid nitrogen coating+gas phase circulation" design: liquid nitrogen only exists in the interlayer of the tank wall and the bottom storage area, and the storage area is a dry deep low-temperature gas phase space. Through natural convection and forced circulation by the top fan, nitrogen is evenly distributed at around -193 ℃, achieving "dry" storage.

2、 Structural analysis: Five fold protection of a "dry" liquid nitrogen tank

1. Double walled structure+vacuum insulation

-Inner wall: 304 stainless steel, corrosion-resistant;

-Outer wall: weather resistant steel+powder coating, salt spray resistant;

-Middle: High vacuum+multi-layer insulation paper, static evaporation rate ≤ 0.8%/day.

2. Nitrogen barrier coating

Liquid nitrogen surrounds the storage area in a "jacket" form, without direct contact with the sample, to avoid the risk of liquid nitrogen backflow.

3. Rotating sample disk

Internally rotatable tray, 2 ml cryovials, 50 ml blood bags, and cube cryovials can all be stored in layers; External handle rotation eliminates the need to probe into the tank, reducing operation time by 50%.

4. Intelligent temperature control system

Dual temperature probes+high and low liquid level sensors, real-time monitoring of temperature difference between the top and bottom; When the temperature is above -180 ℃ or the liquid level is below 30%, an automatic sound and light alarm will be triggered and remote SMS will be sent.

5. Low heating rate

After opening the lid for 10 minutes, the temperature in the storage area only increased by 2 ℃, which is one-third of that of the wet tank, significantly reducing sample freeze-thaw damage.

3、 Core advantages: safety, uniform temperature, large capacity

-Zero liquid nitrogen contact: The storage area is dry, eliminating the hidden dangers of tube bursting and cross contamination;

-Temperature uniformity: The tank wall is coated with liquid nitrogen at 360 ° C, with a temperature difference of ≤ 3 ℃, which is better than that of wet tanks at ≥ 10 ℃;

-Extra large capacity: The V5000EH model can store 42000 2ml cryovials and 1984 50ml blood bags, occupying only 1.4 m ² and increasing capacity density by 30%;

-Operation friendly: One click defogging, visible top cover, detachable console, single person can complete sample loading or sampling in 3 minutes;

-Data compliance: Built in USB and RS-485 interfaces, temperature, liquid level, and lid opening records can be exported to CSV, meeting GMP, ISO 20387, and 21 CFR Part 11 audit tracking requirements.

4、 Application scenario: From "cell bank" to "clinical bedside"

1. Antibody drug development

Primary cells and CHO cell lines need to be stored for a long time, and nitrogen isolation tanks provide a stable environment of -193 ℃.

2. Cell therapy (CAR-T/CAR-NK)

T cells transfected with viral vectors are highly sensitive to temperature fluctuations. Dry storage avoids liquid nitrogen infiltration into the blood bag, reduces virus titer loss, and ensures the efficacy of reinfusion.

3. Umbilical cord blood and stem cell bank

The public repository needs to be stored for more than 20 years, with a low heating rate of the nitrogen isolation tank and dual probe redundancy, meeting the requirements of AABB and NetCord certification.

4. Blood bank and organ bank

HLA typing, lymphocyte and red blood cell deep low-temperature backup, a single can can can store 434 bags of 50 ml blood bags, saving 50% of cold storage space.

5. Gene bank and germplasm resources

Animal embryos, plant seeds, and microbial strains are stored in a dry area to avoid liquid nitrogen infiltration and ice crystal damage, resulting in a recovery rate increase of 10% -15%.

5、 Selection Guide:

1. Capacity calculation: estimated based on "current inventory x 1.3 growth coefficient"; If you plan to use 100000 2ml tubes, you can choose three V5000EH (42000 tubes);

2. Fixed space: door width ≥ 1.2 m, floor height ≥ 2 m, reserved top 0.5 m fan maintenance area;

3. Function selection: If storing CAR-T, Ex db ib IIC T4 explosion-proof electronic control should be selected as an option; If it is a public library, add fingerprint access control and audit tracking software;

4. Check the evaporation rate: Choose models with a static evaporation rate of ≤ 0.8%/day for high temperature and high humidity in the south, and reduce the number of nitrogen supplements;

5. Service comparison: Manufacturers that prioritize providing one-stop services such as "liquid nitrogen supply installation 3Q verification annual calibration" can save 30% of certification time.

6、 Operations and maintenance: making 'safety' a daily routine

1. Daily inspection: Check the temperature and liquid level of the control panel, record abnormal alarms;

2. Weekly maintenance: Blow the sealing strip of the top cover with dry nitrogen to prevent ice residue from affecting the seal;

3. Monthly calibration: Compare the temperature difference between the dual probes using a standard platinum resistor;

4. Annual maintenance: Replace the vacuum nozzle gasket, check the static evaporation rate, and if it exceeds the standard, re evacuate to 1 × 10 ⁻ Pa;

5. Emergency drill: Simulate liquid nitrogen supply interruption every six months, verify the time for the temperature inside the tank to rise from -193 ℃ to -150 ℃, ensure a buffer period, and buy time for sample transfer.
