Do you really understand the culture medium?
Basic culture medium plays a crucial role in organoid culture, and it also needs to be slightly adjusted according to the organoid culture needs of different organ tissue sources. In most organoid cultures, such as gastrointestinal organoids, Advanced DMEM/F-12 is usually chosen as the base medium. However, some literature reports that RPMI-1640 medium is used in the construction of lung organoids, while DMEM medium is used in the construction of brain organoids. This diversity reflects the highly customized nature of organoid culture, providing flexibility for simulating the characteristics of different organ tissues. Therefore, when selecting the basic culture medium, it is necessary to adjust and optimize it according to the specific requirements of the research and the characteristics of the constructed organoids.
Is phenol red necessary in the culture medium?
Phenol red is a commonly used indicator, usually added to the culture medium to determine its acidity and alkalinity, as well as to observe cell growth. The culture medium containing phenol red appears yellow at pH 6.8 and below. When the pH reaches around 7.6, the solution appears pink, and when the pH is above 8.2, it appears purple red. By observing the color change of phenol red, it is convenient for us to determine the pH value of the culture medium.
However, not all cases require the use of media containing phenol red. Phenol red is not an essential component in the culture medium. Many foreign vaccine or antibody manufacturers use phenol red free culture medium in the production process, and the presence of phenol red may even interfere with experimental results. Research has shown that phenol red may have estrogen like activity, meaning that in vitro or in vivo models, it can bind to estrogen receptors and trigger some estrogen reactions, seriously affecting the accuracy and reliability of experimental results. Usually, when we choose matrix gel for organoid culture, we tend to prefer low factor matrix gel without phenol red. However, the existing organoid culture media on the market all contain phenol red, so the role of non phenol red matrix gel is meaningless.

Remove the OC-Advanced-DMEM/F-12 specialized basic culture medium for modeling biological organs from the refrigerator at 4 ° C and restore it to room temperature;
Add 2% O27 medium additive (MB-23061A010, 50X) to the specialized basic culture medium for model based biological organs, so that the final concentration is 1X (for example, 10mL of O27 needs to be added for every 500mL of basic culture medium);
Add different cytokines, small molecule inhibitors, amino acids, hormones, and so on according to the different types of organoids cultured;

Does phenol red have an impact on the culture of cells or organoids?
Research has shown that phenol red has estrogen like functions, meaning that in vitro or in vivo models, it can bind to estrogen receptors and trigger some estrogen responses. In some cell lines (such as breast cancer cells), phenol red can not only promote the proliferation of breast cancer cells, but also promote the expression of estrogen receptor related genes and the activation of estrogen metabolism and signal transduction related pathways in breast cancer cells. In addition, phenol red may cause intracellular sodium/potassium imbalance in serum-free medium, affecting cell growth.
Under what circumstances should phenol red free culture medium be used?
Phenol red is a commonly used pH indicator, usually added to cell culture media to determine the acid-base properties of the medium and observe cell growth. However, some gonadal related cell or organoid cultures may need to avoid using phenol red as it can interfere with experimental results, especially in studies related to estrogen. This is because phenol red itself has a certain estrogenic activity, which may interfere with the estrogenic response of gonadal cells or organoids. Therefore, we recommend using phenol red free culture medium when cultivating cells or organoids related to the gonads.
How can I determine if the nutrient content in the culture medium is sufficient without phenol red?
Organoid culture medium not only provides nutrients for cell growth, but also various cytokines required for organoid growth and differentiation. Therefore, we need to replace the culture medium in a timely manner when cultivating organoids. We usually recommend changing the culture medium every 3 days, which makes the role of phenol red as an acid-base indicator redundant, and phenol red does not have any nutritional components or supportive functions that affect cell growth. Therefore, the presence of phenol red is not necessary in organoid culture.
