The working principle of the fragility tester and its key role in drug quality control
Date: 2025-12-04Read: 1
In the modern pharmaceutical industry, tablets are one of the common and widely used dosage forms. It is not only convenient for transportation, storage, and administration, but also has advantages such as accurate dosage and controllable release. However, tablets are often subjected to mechanical stress during production, packaging, transportation, and use, such as vibration, friction, collision, etc. If the physical strength of the tablet itself is insufficient, it is prone to wear, breakage, or powder loss, which can affect the appearance, uniformity of content, and even efficacy of the drug. Therefore, it is crucial to scientifically evaluate the physical stability of tablets, and the fragility tester is the core equipment used to determine tablet fragility, playing an important role in the drug quality control system.
The working principle is based on simulating the mechanical impact and friction that tablets may encounter in actual circulation processes. Its basic structure usually includes a rotatable drum (usually made of transparent organic glass material) with fixed baffles inside. During testing, a certain number (usually 20 tablets) with a total weight not exceeding 6.5 grams are placed into a drum, and the instrument is set to rotate at a speed of 25 revolutions per minute for 4 minutes, causing the tablets to continuously roll, fall, and collide with each other inside the drum. This process simulates the dynamic stress state of tablets during packaging and transportation. After the test is completed, take out the tablets, remove the surface dust, reweigh them, and evaluate their fragility by calculating the weight loss rate. According to the Chinese Pharmacopoeia and internationally recognized standards, the brittleness and weight loss rate of tablets should generally not exceed 1%, and there should be no breakage, cracking, or crushing.
The key role of the fragmentation tester in drug quality control lies in its objective quantification ability for the physical stability of tablets. Although the hardness of tablets can be measured by a hardness tester, high hardness does not necessarily mean good wear resistance; On the contrary, some soft tablets may also have good resistance to fragmentation. The fragility test comprehensively reflects the overall mechanical tolerance of tablets under dynamic conditions and is an important indicator for evaluating their suitability for industrial production and market circulation.
Secondly, the fragility directly affects the safety and efficacy of drugs. If a large amount of powder is generated during the transportation of tablets due to fragmentation, it will not only cause loss of the main components of the drug, but may also block the holes in the blister packaging, affecting the patient's ability to take the medication; More seriously, mixing powder with other pills may result in inaccurate dosage and pose medication risks. In addition, for coated tablets, excessive brittleness may also damage the integrity of the coating layer, affecting functional characteristics such as sustained-release and enteric coating, thereby altering drug release behavior and interfering with treatment efficacy.
Furthermore, brittle fracture testing is a routine item in drug registration and GMP (Good Manufacturing Practice) inspections. The pharmacopoeias of various countries have set clear standards for tablet fragility. Pharmaceutical companies need to adjust the proportion of prescription excipients and optimize tablet parameters during the process development stage to ensure that their products meet the fragility requirements. In daily production, brittleness is also included in batch inspection items as one of the release criteria to ensure the physical quality consistency of each batch of products.
It is worth mentioning that with the development of pharmaceutical technology, the shredder is also constantly improving. Nowadays, it is equipped with functions such as digital timing, automatic shutdown, dust sealing, etc., which improve the repeatability and operational safety of testing. Some models can also be connected to a data management system to achieve automatic recording and traceability of test results, in line with the trend of digital quality management.
Although the brittle crusher has a simple structure, it plays a crucial role in ensuring the physical quality of tablets. It is not only a testing tool, but also a key bridge connecting prescription design, process control, and end product quality. In today's increasingly strict quality management throughout the entire lifecycle of drugs, scientific and standardized use has profound significance for improving drug quality and ensuring patient medication safety. In the future, with the advancement of intelligent pharmaceuticals and continuous manufacturing technology, fragility testing may be integrated with online monitoring systems, further promoting drug quality control towards real-time and precision.
