How to clean the Thermo Fisher 96 microplate?
Date: 2024-11-04Read: 3
96 well microplate, also known as 96 well cell culture plate or 96 well enzyme-linked immunosorbent assay (ELISA) plate, is a commonly used experimental consumable in laboratories, mainly used for cell culture, drug screening, enzyme-linked immunosorbent assay (ELISA) and other experiments. Due to its large number of pores and small size, cleaning it is relatively complex and requires following certain steps and precautions. Here is toThermo Fisher 96 microplateDetailed introduction of cleaning methods:

1、 Preparation before cleaning

1. Confirm the material of the orifice plate: First, confirm the material of the 96 micro orifice plate, as different materials of orifice plates may require different cleaning methods. Generally speaking, most 96 microplates are made of plastic materials such as polystyrene (PS) or polypropylene (PP).

2. Prepare cleaning tools: Prepare the necessary tools for cleaning, such as pipettes, pipettes, bottles, absorbent paper, etc. Meanwhile, ensure that the cleaning solution (such as PBS buffer or other appropriate solution) has been prepared and placed in the appropriate location.

2、 Cleaning steps

1. Suck out the liquid from the hole: Use a pipette or pipette to suck out the liquid from the hole, being careful not to touch the cells or residue at the bottom of the hole. If there are adherent cells in the well, they can be digested with trypsin, blown into a suspension, and then suctioned out.

2. Add cleaning solution: Use a pipette to add a sufficient amount of cleaning solution to each well, usually around 250 μ l per well. Be careful to slowly add along the wall of the hole to avoid the formation of bubbles.

3. Soak and Shake: Let the cleaning solution soak in the hole for a period of time (such as 3 minutes), then gently shake the hole plate several times to help remove residue. Some laboratories also use shaking tables for oscillation.

4. Suck out cleaning solution: Use a pipette or pipette again to suck out the cleaning solution from the hole, taking care not to touch the bottom of the hole. If necessary, the above cleaning steps can be repeated one or more times.

5. Pat dry or air dry: Place the cleaned Thermo Fisher 96 microplate on absorbent paper and gently pat it several times to remove residual cleaning solution, or place it on a super clean bench to air dry. Be careful not to use paper towels to wipe the bottom of the orifice plate to avoid affecting the reading.

3、 Precautions

1. Avoid cross contamination: Pay attention to avoiding cross contamination during the cleaning process, especially when handling multiple different samples. It is recommended to process only one sample of the orifice plate at a time and replace the pipette tip and gloves with new ones after processing one sample.

2. Control the cleaning force: During the cleaning process, pay attention to controlling the force to avoid excessive force that may cause damage to the orifice plate or cell detachment.

3. Choose the appropriate cleaning solution: Select the appropriate cleaning solution according to the experimental needs, and ensure that its pH value and ion concentration are suitable for cell growth or subsequent experiments.

The cleaning of the Thermo Fisher 96 microplate is a process that requires careful and patient attention. By following the correct steps and precautions, the cleaning effect can be ensured and the service life of the microplate can be extended.
