Arsenic and its compounds can be absorbed into the human body through the respiratory tract, digestive tract, and skin. 95% -99% of arsenic in the blood binds to globin in red blood cells. Arsenic in tissues is mainly distributed in the liver, kidneys, gastrointestinal walls, muscles, and other areas. Skin, hair, nails, and bones can serve as strong storage reservoirs for arsenic. Arsenic in the body is mainly excreted by the kidneys and digestive tract, with some being excreted by the skin, hair, and nails. The toxic effect of arsenic is that arsenic ions bind with thiol and hydroxyl groups in the molecular structure of enzyme proteins in the body, causing the enzyme to lose activity. Inhibition of pyruvate oxidase, choline oxidase, transaminase, alpha glycerophosphate dehydrogenase, 6-phosphate glucose dehydrogenase, or cytochrome oxidase can interfere with the normal metabolism of cells, affect respiration and oxidation processes, cause cell pathology, and inhibit cell division and proliferation. In addition, arsenic acid and arsenite acid can replace phosphate in many biochemical processes, thereby uncoupling the oxidative phosphate process, reducing the formation of high-energy phosphate bonds, and interfering with cellular energy metabolism. Metabolic disorders can first harm nerve cells, causing symptoms of toxic neurasthenia and multiple neuropathy. Arsenic can directly damage the walls of small arteries and capillaries, as well as act on the vasomotor center, causing smooth muscle paralysis and increased permeability of the vascular wall, leading to a decrease in blood volume and exacerbating organ damage. Arsenic sulfide such as realgar and orpiment have low solubility and toxicity in water; Arsenic trioxide has high water solubility and is also highly toxic. Arsenic trioxide and arsenic trichloride have irritating effects on the eyes, upper respiratory tract, and skin.
The mechanism of arsine poisoning is different: inhibiting the action of glutathione peroxidase leads to the formation of peroxides and hemolysis; It is also possible that arsenic binds to thiol groups, damaging the sodium and potassium pump function of red blood cell membranes. After extensive intravascular hemolysis, acute renal failure often occurs. The dose of arsenic poisoning is 5-50mg, and the lethal dose is 60-200mg.
