Sartoris pipette: bubble free technique for culture medium transfer
Date: 2025-12-24Read: 2

The cultivation of genes contains components such as serum and protein, which can easily generate bubbles when transferred, affecting the accuracy of cell culture and experiments.
Sartoris pipettes (such as Tacta) ® Manual Picus ® 2 electric series) combined with dedicated suction heads and standardized operation,
Zero bubble generation can be achieved in rapid liquid transfer. The following technical solutions are provided from four aspects: selection, technique, parameters, and maintenance.

1、 Pre preparation: Accurate matching of equipment and consumables

• Selection of pipettes:
Select the appropriate range based on the pipetting volume, and it is recommended that the pipetting volume be ≥ 30% of the nominal capacity of the pipette. For example, for a volume of 1000 μ L, the 1000 μ L specification is preferred,
To avoid operating instability and bubbles caused by overloading of small processes. Electric version (Picus) ® 2) Can lock the suction and discharge speed for better consistency.

• Suction head matching:
Using the original suction head from Sartoris, with Optiload ® Spring cone installation, lightly press into place to seal, prevent air leakage and intake;
Wide mouthed suction tips can be used to remove viscous culture media, reducing liquid flow resistance.

Pre lubrication and exhaust:
Before formal pipetting, pre moisten the pipette tip 3-5 times with the target culture medium to fully saturate the inner wall of the pipette tip, remove residual air, and saturate the air column inside the pipette tip to reduce bubble generation.

2、 Core operation: Reverse pipetting+speed and angle control, fast and bubble free

The reverse pipetting method of the Sartorius pipette is the key to preventing bubbles, and when combined with smooth operation, it can balance speed and no bubbles. The steps are as follows:

1. Liquid absorption:
First press to the second gear (liquid discharge gear), vertically immerse the suction head 2-3mm below the liquid level of the culture medium, slowly release the button to suck the liquid, hold for 1-2 seconds after sucking, and remove it after the liquid is stable.

2. Drainage:
Place the suction head at a 30-45 ° angle against the inner wall of the container, slowly press it to the suction mode, and let the liquid flow down along the wall; The remaining liquid should be left inside the suction head and not blown out to avoid the formation of bubbles due to airflow impact.

3. Speed up tips for electric models:
Picus ® 2. Low flow rate gears (such as 1-2 gears) can be set, and the multi liquid dispensing mode can be enabled to dispense multiple times at once, reducing the bubbles caused by repeated suction and discharge;
Manual version with the help of Tacta ® The labor-saving design maintains a constant suction and discharge speed, and the thumb movement is smooth without any jerks.

4. Detail control:

When suctioning, move the suction head down synchronously with the decrease of the liquid level, always keeping the suction head below the liquid level to avoid inhaling air; To avoid vertical impact on the liquid level during drainage, there should be no violent movements throughout the entire process.

3、 Advanced Optimization: Sample and Equipment Parameter Adaptation

• Sample processing:
If there are bubbles in the culture medium, let it stand for 5-10 minutes or centrifuge at low speed (1000rpm, 5 seconds) to remove the bubbles before removing it; Avoid vigorous shaking of the culture medium to reduce bubble formation.

• Parameter calibration:
Regularly calibrate the pipette according to specifications to ensure accurate capacity; Install a safety cone filter to prevent liquid backflow from contaminating the instrument, while ensuring stable airflow and reducing the risk of air bubbles.

4、 Daily maintenance: ensuring long-term stability and anti foaming

After each use, wipe the lower half of the pipette and the tip cone with 70% ethanol to prevent residual culture medium from contaminating and affecting the seal;

Optiject is used to remove the suction head ® Soft lever, avoid hitting the pipette;

Regularly check the sealing of the suction head and the smoothness of the piston, and promptly repair and calibrate in case of jamming or leakage.

5、 Conclusion: Technology empowers efficient experiments

The Sartoris pipette features reverse pipetting, precise speed control, and ergonomic design, combined with standardized operation,
It can achieve rapid transfer of culture medium without bubbles, suitable for scenarios such as cell culture and biopharmaceuticals, and help improve experimental efficiency and result stability.
