Installation and Environmental Requirements for Atomic Fluorescence Spectrometer Equipment
Date: 2025-10-27Read: 2
Atomic Fluorescence Spectrometer (AFS) is a highly sensitive analytical instrument widely used in environmental monitoring, food safety testing, geological and mineral analysis, and other fields. Its management and use require strict adherence to standardized procedures to ensure the accuracy of data, long-term stability of equipment, and safety of operators.
1、 Installation and Environmental Requirements for Atomic Fluorescence Spectrometer Equipment
1. Laboratory layout planning
Choose a location away from vibration sources (such as centrifuges), electromagnetic interference sources (large motors), and direct sunlight to install the instrument. The workbench should be sturdy and stable, with a load-bearing capacity that meets the weight requirements of the instrument. Maintain good ventilation in the laboratory to avoid the accumulation of corrosive gases that may affect the performance of optical components.
Reserve sufficient operating space for daily maintenance and sample replacement, and set up a dedicated area to store reagents and standards to avoid cross contamination.
2. Temperature and humidity control standards
Maintain room temperature within the range of 15-30 ℃ and humidity between 40% and 70% RH. Excessive humidity may cause circuit short circuits or mold growth in optical components, while low humidity can easily generate electrostatic interference in signal acquisition. It is recommended to install an air conditioning system and a dehumidification device, and regularly monitor and record changes in environmental parameters.
3. Equipping with safety protection facilities
Install eye wash stations and emergency sprinkler systems in nearby locations to respond to chemical sputtering accidents; Fire extinguishing equipment shall be provided (carbon dioxide type is preferred), and foam fire extinguishers shall not be used to avoid damage to precision parts. The experimental table is equipped with corrosion-resistant tray liners to prevent acid leakage from corroding the equipment casing.
2、 Daily operation specifications for atomic fluorescence spectrometer
1. Preparation before startup
According to the "Instrument and Equipment Usage Registration Form", check the maintenance status after the last use, confirm that the pressure of the argon gas cylinder is sufficient (usually ≥ 0.5MPa), the liquid level of the carrier fluid is normal, and there is no sediment. Check if there is any looseness or leakage at the connections of each pipeline, especially the integrity of the sealing ring of the hydride generation system.
Preheat the stabilized power supply for 30 minutes in advance to achieve stable output of light source intensity. Load the standard curve solution required for the day for baseline calibration, ensuring that the instrument is in test mode rather than energy optimized state.
2. Sample pre-treatment techniques
Solid samples need to be subjected to microwave digestion or acid leaching followed by volumetric filtration to remove suspended particles; Take an appropriate amount of liquid sample and add borohydride * reducing agent directly to initiate hydrogenation reaction. Pay attention to maintaining consistent acidity of the sample medium (usually 5% hydrochloric acid medium) to avoid fluorescence quenching effects caused by pH differences.
Prepare blank control samples using ultrapure water to reduce background interference. For complex matrix samples, co precipitation method can be used to enrich the target elements before testing on the machine.
3. Key points for monitoring the measurement process
When using the standard addition method to draw the working curve, at least 5 concentration gradient points should be set, and each point should be measured in parallel three times and the average value should be taken. Real time observation of the fluctuation range of fluorescence signal response values, if RSD>5%, re injection analysis is required. The needle of the automatic sampler should be rinsed with ethanol after each cleaning to prevent cross contamination.
After each batch of samples is tested, the system flushing procedure is executed. First, residual reagents in the pipeline are flushed with deionized water, and then diluted nitric acid is injected into the reaction tank for 10 minutes to decompose metal hydroxide deposits.
