3D printing integrated microfluidic optogenetic neural probe, integrating light stimulation and drug/virus delivery functions
Date: 2025-11-24Read: 2
Optogenetics technology, with its high-precision regulation ability of neuronal activity, has become an important tool in modern neuroscience research. Traditional optogenetics experiments require two independent surgeries: injection of a virus vector carrying the photosensitive protein gene, followed by a second surgery to implant a light stimulation probe after 1-2 weeks of gene expression. This dual surgical approach not only increases experimental complexity, but also exacerbates tissue damage and inflammatory response.

Recently, a research team from New York University Abu Dhabi has developed a3D printed multimodal optogenetic neural probe (MIO),Integrating optogenetic stimulation with microfluidic drug/virus delivery functionality into a single deviceThis study successfully solved the problem of multiple surgeries required in traditional optogenetics research, providing an efficient, customizable, and minimally invasive solution for the field of neuroscience. Accurate delivery of viral vectors or drugs. This design breakthrough optimizes the traditional dual surgical process of virus injection and probe implantation into a single operation, significantly simplifying the experimental steps. Researchers successfully implanted probes in the subthalamic nucleus (STN) of mice, a key brain region for motor regulation, and completed virus vector injection and device fixation simultaneously through a single surgery (Figure 1).Figure 1. (a) The traditional process involves stereotactic injection of viral vectors to express photosensitive opsin in target neurons, followed by surgical implantation of optical probes to deliver controlled light pulses for neuronal modulation. (b) The integrated process of implanting optogenetic devices for virus transmission combined with microfluidic control. It involves stereotactic placement of the device in a target brain area, where microfluidic control delivers viral vectors to express photosensitive opsins in specific neurons, followed by simultaneous or subsequent activation of optical probes to deliver controlled light pulses for precise neuronal modulation. In terms of performance characterization, the research team focuses on MIO

probeSystematic testing was conducted (Figure 2). The electrical and optical test data (Figure 3) show that the device can stably output blue light stimulation at a working voltage of 3.0V (corresponding to a current of about 12mA), and its peak emission wavelength (465nm) highly matches the response spectrum of ChR2 photosensitive protein, with light intensity exceeding the activation threshold (1mW/mm ²).

Thermal validation experiments have shown that under simulated in vivo environmental conditions, even with higher stimulation frequencies and pulse widths, the temperature rise of the probe can be strictly controlled within the safe threshold of 2 ° C. This result confirms that the probe has excellent thermal management capabilities, which can effectively avoid thermal damage to surrounding nerve tissue and meet the stability requirements of physical properties for long-term in vivo neural regulation.

The study systematically compared the experimental group implanted with MIO probes with the control group using traditional fiber optic probes (requiring two surgeries) through a seven week behavioral experiment (Figure 5). The data shows that optogenetic stimulation can significantly improve the movement distance and speed of both groups of mice, proving that both devices can effectively activate STN neurons and regulate motor behavior. It is worth noting that the MIO probe group exhibited more stable and consistent behavioral responses throughout the entire experimental cycle, suggesting that the integrated implantation strategy of a single surgery may endow the device with better long-term stability and regulatory effects.

The biocompatibility and functional effectiveness of the MIO probe were further validated through immunohistochemical analysis using mCherry fluorescence labeling. The researchers observed significant mCherry expression in the STN region, indicating that the MIO probe successfully achieved targeted delivery of viral vectors and guided efficient expression of ChR2 protein in target neurons. This result confirms the feasibility of a single surgical integration scheme at the molecular level, providing key evidence for the application of the device in long-term neural regulation (Figure 6).Figure 6. Representative horizontal slices of mCherry expression near the implantation trajectory in the STN region.

Compared with the control group, the density of neurons around the MIO probe implantation area was significantly higher. Quantitative analysis using NeuN staining showed that the MIO group exhibited better neuronal preservation in various regions 50-200 μ m away from the implantation site (Figure 7).

图片
Immunohistochemical analysis of the astrocyte marker GFAP showed a significant decrease in the activation level of astrocytes around the implantation site in the MIO group (Figure 8). At the same time, the number of activated microglia labeled with ED1 also significantly decreased, confirming the effective control of inflammatory response (Figure 9).

This series of histological evidence strongly confirms the unique advantages of a single surgical strategy: by avoiding repeated tissue trauma, it significantly reduces mechanical damage and secondary inflammatory reactions caused by implants. This protective effect is not only reflected in the preservation of the number of neurons, but also in the reduction of glial scar formation, providing a solid histological foundation for the long-term functional stability of the device.
