Packing is the core of HPLC column preparation
Date: 2025-06-18Read: 1
HPLC column preparation is an important tool for separating and purifying compounds in High Performance Liquid Chromatography technology.
-Column tube: usually made of stainless steel or glass, with good pressure resistance and chemical stability. The inner diameter of the column determines the loading capacity and separation scale of the chromatography column. Generally, the inner diameter of the prepared chromatography column is in the range of 50-200mm.
-Packing: It is the core part of the chromatographic column, and common packing materials include silica gel, alumina, organic polymer microspheres, etc. The filler of silicone matrix has good mechanical strength and chemical stability. By bonding different functional groups (such as C18, C8, C4, etc.), it can achieve the separation of different polar compounds.
-Sieve plate: located at both ends of the column tube, used to support the packing, prevent packing leakage, and ensure that the mobile phase can pass evenly through the chromatographic column.
Steps for using HPLC to prepare chromatographic columns:
1. Preparation work
-Safety check: Ensure the safety of the laboratory area, wear personal protective equipment such as laboratory clothing, gloves, and goggles.
-Instrument inspection: Check the status of HPLC equipment to ensure that components such as columns, detectors, pumps, etc. are operating normally. Open the HPLC system and set appropriate parameters such as temperature and flow rate to achieve a stable state of the system.
-Mobile phase preparation: Prepare the mobile phase according to experimental requirements, filter the solvent or mobile phase through 0.2 μ m filter paper, and perform ultrasonic degassing to remove bubbles and impurities.
-Sample preparation: Dissolve the sample to be tested in an appropriate solvent and dilute it to the appropriate concentration. Filter it through a 0.2 μ m filter membrane to ensure that the sample solution is clear and free of impurities.
2. Install chromatography column: Install the prepared chromatography column in the HPLC system, paying attention to tight connections to prevent liquid leakage. Before installation, check the appearance of the chromatography column for any damage or other issues.
3. System equilibrium: Rinse the chromatography column with mobile phase for 15-30 minutes until the baseline is stable, to ensure that the column is filled with mobile phase and reaches equilibrium.
4. Injection determination: Select the appropriate injection method and injection volume, and inject the sample into the HPLC system. Start collecting data and record information such as retention time, peak width, and peak height of chromatographic peaks.
5. Data processing and analysis: Based on the recorded data, calculate the relevant parameters of the chromatographic column, such as theoretical plate number, asymmetry factor, separation degree, etc., to evaluate the performance of the chromatographic column. Common data processing methods include peak area measurement, peak height measurement, quantitative analysis, and qualitative analysis.
6. Cleaning and preservation of chromatography column: After the measurement is completed, clean the chromatography column with an appropriate solvent to remove residual samples and mobile phase. For reverse phase chromatography columns, they are usually washed with methanol or acetonitrile first, and then with pure water; For normal phase chromatography columns, organic solvents such as n-hexane can be used for flushing. After cleaning, store the chromatography column properly to avoid drying and contamination.
