Oligosaccharide separation packing materialEfficient separation is achieved through multidimensional physical and chemical interactions.Oligosaccharide separation packing materialThe core mechanism includes:
1. Molecular sieving:
The screening effect of molecular size based on porous structure. For example, the polyacrylamide gel (Bio Gel P22/P24) restricts the penetration rate of oligosaccharides through the pore size to achieve the separation of maltooligosaccharides, dextrose oligosaccharides, etc. according to the degree of polymerization. Bio Gel P24 can clearly distinguish between disaccharide and hexose components when separating oligosaccharides under column temperature of 60 ℃ and flow rate of 55mL/h.
2. Affinity adsorption:
The chemical groups on the surface of the filler form specific interactions with oligosaccharide molecules. Activated carbon diatomaceous earth composite fillers can efficiently separate the trisaccharide to hexose components in the enzymatic hydrolysis products of lactitol by hydrophobic interaction and van der Waals adsorption of oligosaccharides, combined with ethanol gradient elution (linearly increasing concentration from 0-25%).
3. Ion exchange:
Charged fillers selectively adsorb oligosaccharides through electrostatic interactions. Calcium based sulfonic acid resin (cation exchange) utilizes the difference in binding strength between fructose and glucose on the resin to achieve industrial scale separation, with a single cycle processing capacity of up to tons and fructose purity exceeding 95%.
4. Gel penetration:
Separation mechanism based on molecular volume differences. Cellulose fillers separate neutral sugars through the exclusion effect. In the water saturated n-butanol isopropanol system, oligosaccharides with arabinose groups in the enzymatic hydrolysis products of arabinoxylan achieve baseline separation from linear xylan.
