The Nikon EI Biological Microscope is a high-precision microscope used in biological research and medical fields, widely used in cytology, histology, microbiology, and other fields. The operation steps usually include preparing samples, adjusting microscopes, observing and recording images, etc. The following are the basic operation steps:
1. Preparation work
a. Check the equipment
Ensure that the power supply, light source, and other accessories of the microscope are intact.
Clean the lens and sample stage to avoid dust and pollution affecting the observation effect.
Check if the connection wires of the microscope (such as power cord, camera connection wire, etc.) are normal.
b. Sample preparation
Prepare suitable samples according to research needs, such as slices, cultured cells, blood smears, etc.
If slides and coverslips are needed, make sure they are clean and free of bubbles or impurities.
Place the sample on the stage and gently secure it with tweezers or other tools.
2. Basic operation of microscope
a. Power on
Turn on the power switch of the microscope and ensure that the light source is turned on.
If the microscope is equipped with an LED light source or halogen lamp, adjust the brightness of the light source as needed.
b. Install eyepiece and objective lens
Check the selection of eyepiece and objective lenses to ensure they are suitable for the research purpose.
Generally speaking, objective lenses are divided into low magnification, medium magnification, and high magnification. When starting observation, it is usually done with a low magnification objective lens (such as 4x or 10x).
c. Adjust the light source
Adjust aperture, brightness, and contrast to achieve appropriate lighting conditions.
When using phase difference or fluorescence observation, ensure that the corresponding filter or fluorescence module has been installed.
3. Sample observation
a. Coarse adjustment of focal length
Rotate the objective lens to a low magnification position and use the coarse focus knob to adjust the sample to make it roughly clear.
Adjust the height of the stage as needed to align the sample with the objective lens.
b. Fine tuning focal length
Use the fine focus knob for precise focusing to ensure clear images. At this point, you can zoom in and gradually switch to a higher magnification objective lens (such as 40x, 100x).
c. Switch objective lens
After seeing the area of the sample under the low magnification objective, gradually switch to the medium or high magnification objective for more detailed observation.
When switching objectives, do not directly rotate the objective lens with force, but first slightly raise and lower the stage to avoid damaging the objective lens and sample.
d. Adjust the field of view
Use the focusing ring or distance adjustment of the eyepiece to ensure a clear and comfortable field of view.
If necessary, the horizontal and vertical positions of the sample stage can be adjusted to ensure the accuracy of the observation area.
4. Image recording and analysis
a. Image acquisition
If the microscope is equipped with a camera or digital imaging system, image acquisition can be performed through the software provided with the microscope.
Choose the appropriate shooting resolution and exposure time to ensure image quality.
b. Image saving and analysis
Save the collected images to the computer for later analysis.
Use the software provided with the microscope for image enhancement, contrast adjustment, and other operations to further improve the readability and analysis effectiveness of the images.
5. End the operation
a. Turn off the device
After observation, first adjust the objective lens back to the low magnification position.
Turn off the light source and power off the microscope to prevent prolonged use of the light source from affecting its lifespan.
b. Cleaning microscope
Gently wipe the eyepiece and objective lens with lens paper or cleaning cloth to avoid leaving fingerprints or stains.
Clean the sample stage, stage, and other components to maintain the hygiene of the microscope.
c. Storage microscope
If the microscope is no longer in use, it is best to cover it with a dust cover to avoid the accumulation of dust or impurities.
Ensure that the microscope is stored in a dry and warm place, avoiding prolonged exposure to strong light or humid environments.
6. Special precautions
When using a high-power objective lens (such as an oil lens), lens oil (such as immersion oil) must be used to improve image clarity and prevent lens damage.
When using fluorescence microscope mode, it is important to ensure the matching of lighting and filters to achieve the best fluorescence effect.
Regularly calibrate the microscope to ensure its accuracy and stability.
