Sharing of Scientific Maintenance Methods for Biotage Rapid Preparation of Liquid Chromatography Instruments
Date: 2025-10-20Read: 1

Biotage rapid preparation liquid chromatography is a core equipment in the fields of drug development, natural product separation, and high-purity compound purification. With its high flow rate, large sample loading capacity, and automation control capabilities, it can efficiently achieve rapid separation and enrichment of target components. Its system is complex, including key components such as pumps, injectors, chromatography columns, detectors, and fraction collectors. It is susceptible to solvent corrosion, sample residue, and particle blockage during long-term operation. The scientific maintenance and upkeep of biotage rapid preparation liquid chromatography instruments are key to ensuring separation efficiency, extending equipment lifespan, and ensuring purification quality.

1、 Daily maintenance
After each run, execute the 'cleaning program': rinse the entire flow path with an appropriate solvent (such as a water methanol gradient) for at least 30 minutes to remove residual buffer salts and organic matter. Turn off the detector light source to prevent unnecessary loss of lamp life. Empty the waste liquid bottle, wipe the surface of the instrument, and keep it clean.
2、 Weekly maintenance
Pump system: Check if the solvent filter head is clogged, clean or replace it with ultrasonic waves. Observe whether there is any leakage in the pump head and whether the sealing ring is aging.
Automatic sampler: Clean the outer wall and needle holder of the injection needle to prevent cross contamination of the sample. Check if the quantitative loop and flow path are unobstructed.
Detector: Gently wipe the flow cell window with a dust-free cloth to avoid scratches. Check baseline noise and drift, and perform baseline correction if necessary.
3、 Monthly maintenance
Reverse wash the chromatographic column (if allowed) to remove column head contaminants. Use specialized cleaning solution or high proportion organic phase flushing system. Check if the pipeline joints are loose and replace the aging PEEK or stainless steel pipes. Test the flow accuracy and pressure stability of the pump to ensure accurate delivery.
4、 Quarterly maintenance
Replace vulnerable parts such as the active valve and sealing ring of the pump. Clean or replace the online filter (0.45 μ m). Perform wavelength calibration (using standard filters) and sensitivity verification on the detector. Check if the mechanical arm of the fraction collector moves smoothly and its position is accurate.
5、 Maintenance every six months
Conduct system suitability testing using standard samples (such as benzoic acid) to evaluate key parameters such as separation degree, theoretical tray number, and repeatability. Conduct a comprehensive inspection of electrical connections, grounding conditions, and cooling systems (such as column temperature box fans). Backup method files and system settings.
