  Yeast micro manipulatorThe operating steps and key points of the stage are as follows:
1、 Preparation before operation
Equipment inspection
Confirm that the stage of the microscope operator is clean and free of debris, and that the culture dish or slide is securely placed to avoid sliding during operation.
Check the connection status between the stage, microscope light source, and microscope manipulator to ensure the normal operation of the equipment.
Sample preparation
Yeast cell samples need to be prepared in advance and placed in a suitable culture environment (such as culture dishes or glass slides) to ensure cell viability and observation status.
If staining observation is required, iodine solution or other staining agents can be added dropwise, and absorbent paper can be used to attract from the other side to achieve uniform staining.
2、 Operating steps of the loading platform
Place the sample
Place the prepared yeast cell sample (such as a glass slide or culture dish) face up on the stage.
Use a compression clamp to secure the sample, ensuring it is stable and not moving, and avoiding damage to optical components or affecting observation accuracy during operation.
adjust the position
By using the controller of the electric stage (such as a joystick or touch screen), move the stage so that the sample is facing the center of the through-hole.
If you need to observe a specific area, you can slowly move the stage to find the best focusing plane.
Focusing observation
Rotate the coarse focus spiral of the microscope to slowly lower the barrel, while observing from the side to avoid the objective lens crushing the sample.
With one eye fixed on the eyepiece, rotate the coarse focus screw to raise the lens barrel until an object image appears in the field of view.
Fine tune the fine focus spiral to make the object image clearer. If the object image is offset, the position of the stage or the direction of the sample can be fine tuned.
Automated operations (if applicable)
If the device supports automated processes such as tetrad dissection and single-cell screening, it can initiate automated operations by setting parameters through built-in software.
Maintain stable technique during operation to avoid affecting accuracy due to shaking.
3、 Post operation maintenance
Cleaning and organization
After the experiment is completed, turn off the microscope manipulator and microscope light source, and clean the lens and workbench with specialized lens cleaning paper.
Store the device in a dry environment and cover it with a dust cover when not in use for a long time to prevent damage to the optical components.
regular maintenance
Regularly lubricate the robotic arm and positioning device of the micro manipulator to ensure smooth movement.
Check if all connecting components are loose and tighten them in a timely manner to avoid a decrease in accuracy.
Regularly calibrate the positioning accuracy of the stage to ensure the accuracy of experimental data (recommended to be operated by professionals or guided by the instructions).
4、 Precautions
Avoid excessive squeezing
Avoid excessive squeezing or damaging yeast cells during operation to ensure that the sample is in a better observation state.
Prevent vibration interference
The equipment should be placed in an environment with suitable temperature and humidity, avoiding vibration and direct sunlight to avoid affecting the observation effect.
Standardize operational procedures
Strictly follow the operating instructions for the experiment to avoid equipment damage or experimental failure due to improper operation.
