The widespread application of shaker incubators in laboratories
Date: 2025-05-27Read: 3
The shaker incubator is an important equipment in laboratories that integrates oscillation cultivation and constant temperature control. With its flexible control of temperature, rotation speed, and time, it plays a key role in microbial cultivation, cell biology, molecular biology, biopharmaceuticals, and other fields.

The following will introduce the core functions, application scenarios, operation points, and maintenance suggestions:
1、 Core functions and technical features
1. Multi functional integrated design
Constant temperature control: using microcomputer PID temperature control technology, the temperature range is usually 4 ℃ (with refrigeration function)~60 ℃, and the accuracy can reach ± 0.1 ℃, meeting the strict requirements of different biological samples for cultivation temperature (such as Escherichia coli suitable at 37 ℃, yeast suitable at 28 ℃).
Oscillatory culture: Reciprocating or rotating oscillation is achieved through eccentric wheels or magnetic drive, with a speed range of generally 20~300 rpm and an amplitude of mostly 20~50 mm, ensuring sufficient contact between the culture medium and air to promote the growth and metabolism of cells or microorganisms.
Intelligent control: supports timing function (0-999 hours), real-time display of speed/temperature, and over temperature alarm.
2. Structure and safety
Material: The inner liner is mostly made of stainless steel or engineering plastic, which is corrosion-resistant and easy to clean; The shell is made of cold-rolled steel plate sprayed with plastic, which has good insulation and sound insulation effect.
Capacity: According to the tray size, it is divided into small (accommodating 2-4 500 mL shake flasks), medium (accommodating 8-12 2 L shake flasks), and large (accommodating 20 or more 5 L shake flasks), suitable for different experimental scales.
Safety design: Equipped with overload protection, leakage protection, and door lock devices to avoid safety hazards caused by sample spillage or equipment failure during oscillation.
2、 Typical application scenarios in the laboratory
1. Microbial cultivation and fermentation
Strain propagation: used for liquid medium oscillation cultivation of bacteria (such as Escherichia coli) and fungi (such as yeast), increasing dissolved oxygen content through high-speed oscillation, shortening logarithmic growth period, and improving strain yield. For example, in genetic engineering experiments, a shaker incubator can rapidly amplify engineered bacteria containing target plasmids.
Fermentation process optimization: By adjusting the rotational speed (affecting dissolved oxygen) and temperature (affecting enzyme activity), the optimal conditions for microbial fermentation are screened. The fermentation of penicillin in industrial production requires cultivation at 25 ℃ and 150 rpm for 7-10 days.
2. Cell biology research
Wall attached cell culture: Paired with specialized cell culture bottles (such as T75 cell culture bottles), low-speed oscillation (50-80 rpm) is used to prevent cell aggregation and promote uniform distribution of nutrients in the culture medium, suitable for the cultivation of fibroblasts, liver cells, etc.
Suspension cell culture: For large-scale cultivation of CHO cells (Chinese hamster ovary cells) and HEK293 cells, shaking at 37 ℃ and 120 rpm is required to ensure that the cells are suspended and fully exposed to oxygen and nutrients.
3. Molecular Biology Experiment
Plasmid extraction and purification: After overnight cultivation of bacteria containing plasmids on a shaker, the bacterial cells can be collected by centrifugation for further extraction of plasmid DNA. For example, the pUC series plasmids commonly used in the laboratory need to be cultured in LB medium at 37 ℃ and 220 rpm for 12 hours before extraction.
Protein expression and induction: When using IPTG to induce the expression of recombinant proteins in Escherichia coli, it is necessary to first shake culture at 37 ° C until the logarithmic growth phase, and then cool down to 20 ° C~25 ° C to induce expression, in order to avoid the formation of inclusion bodies.
4. Biopharmaceuticals and vaccine production
Virus culture: When cultivating rod-shaped viruses in insect cells (such as Sf9 cells), it is necessary to shake the culture at 27 ℃ and 100 rpm to ensure efficient replication of virus particles.
Preparation of monoclonal antibodies: Suspension culture of hybridoma cells should be carried out at 37 ℃, 5% CO ₂ (supported by some models), and 80 rpm. A constant temperature and humidity environment in a shaker incubator can improve cell survival rate and antibody production.
5. Environmental Science and Food Testing
Microbial detection in water quality: used for shaking culture of diluted water samples. For example, when measuring biochemical oxygen demand (BOD ₅), it is necessary to cultivate for 5 days at 20 ℃ and 120 rpm to simulate the natural decomposition process of water bodies.
Food microbiological testing: In the detection of total bacterial count, coliform bacteria, etc., the shaking incubator can quickly activate the microorganisms in the sample and shorten the detection cycle.
