Inhalation preparations, as the core dosage form for treating respiratory diseases such as asthma and chronic obstructive pulmonary disease (COPD), are highly dependent on the drug's dissolution and absorption process in the lungs for their efficacy. The inhalation formulation dissolution module is a key technology designed to crack this complex mechanism. By simulating the human lung environment and accurately quantifying the dissolution behavior of aerosol particles, it has become a core tool for drug development and quality control.
1、 Technical principle: In vitro reconstruction of lung microenvironment
The inhalation formulation dissolution module works in conjunction with an aerosol generator to construct an artificial blood and blood barrier in vitro. The system can accurately control experimental parameters: temperature fluctuation range ± 0.5 ℃, humidity accuracy ± 2% RH, and simulate aerosol deposition under different breathing modes (0.1-5 μ L/cm ²). Taking the DissolvIt module as an example, its core components include:
1. Dissolution chamber: made of polytetrafluoroethylene material to avoid drug adsorption;
2. Inverted microscope: real-time observation of particle dissolution process, with a resolution of 0.5 μ m;
3. Dissolution medium circulation system: supports simulated lung fluid containing lung surfactants (such as DPPC), with a pH adjustable range of 4.5-7.4.
Experimental data shows that the module has a dissolution detection error of less than 5% for poorly soluble drugs, which is three times more accurate than the traditional flow cell method.
2、 Application value: Empowering the entire chain from research and development to quality control
During the drug development phase, the dissolution module can generate pharmacokinetic (PK) curves, with the CV values of key parameters Cmax (peak blood concentration) and Tmax (time to peak) being less than 10%. For example, after optimizing the prescription of a new asthma drug through this module, the lung deposition rate increased from 32% to 48%, and the bioavailability increased by 1.8 times. In the consistency evaluation of generic drugs, if the similarity factor f2 of the dissolution curve is greater than 50, partial in vivo experiments can be exempted, significantly shortening the research and development cycle. In addition, this module can also be used to evaluate the performance of inhalation devices. For example, a certain aerosol was tested and found to have valve leakage causing delayed dissolution. After improvement, the dissolution time was reduced by 40%.

3、 Industry Challenges and Future Directions
Despite mature technology, the dissolution module still faces three major challenges:
1. Individual difference simulation: It is necessary to integrate variables such as respiratory flow rate and pulmonary surfactant composition;
2. Multiphase system treatment: For suspended formulations, it is necessary to optimize the design of the stirring blade to reduce particle agglomeration;
3. Data standardization: The industry has not yet established a unified formula and evaluation method for dissolution media.
In the future, with the introduction of AI algorithms, the dissolution module will achieve automatic parameter optimization and intelligent interpretation of results, combined with microfluidic technology to further reduce device size and promote the development of inhalation formulations towards precision and portability.
