In the fields of biomedicine, nanomaterials, food and beverage, cosmetics, and chemical engineering, achieving high uniformity of materials at the microscale is the key to improving product performance, stability, and bioavailability. Although traditional high-pressure homogenizers have been widely used, they often face problems such as wide particle size distribution, poor batch repeatability, and shear thermal damage when processing highly sensitive systems such as nanoparticles, liposomes, and mRNA vaccines. The microjet homogenizer, with its unique fixed geometry microchannel design, ultra-high pressure, and controllable shear force, has become an equipment for achieving sub micron and even nanometer level homogenization, emulsification, de aggregation, and cell fragmentation, and is known as the "precision engine of nanomanufacturing". This article will comprehensively analyze the core competitiveness of this homogeneous equipment from its working principle, technological advantages, typical applications, and industry value.
1、 What is a microjet homogenizer?
Microfluidic homogenizer is a high-pressure homogenizer device that uses a fixed microchannel interaction chamber. The core lies in the fact that materials are driven at ultra-high pressure (usually 5000-30000 psi, or 35-200 MPa) and pass through micrometer sized Y-shaped or Z-shaped cross channels at high speed, where they undergo quadruple physical effects of impact, high-frequency shear, cavitation effect, and turbulent disturbance, thereby achieving efficient particle crushing and uniform dispersion.
Unlike traditional valve homogenizers such as APV and Gaulin, which rely on adjustable valve clearances, the interaction chamber of micro jet homogenizers is made of precision sintered ceramics or diamond composite materials, with a fixed geometric structure to ensure that the fluid passing through each time experiences the same energy input, thereby achieving process reproducibility and particle size control accuracy.
2、 Core working principle and technical characteristics
1. The synergistic effect of the quadruple homogenization mechanism
Collisions: Two high-speed fluids collide head-on at the center of a microchannel, instantly converting kinetic energy into fracture energy;
High shear: shear rate generated when flowing through a narrow channel (diameter 50-200 μ m);
Hole effect: Local pressure drops sharply, forming bubbles and collapsing, producing micro jet impact;
Turbulent disturbance: Reynolds number at the microscale, enhancing mixing and dispersion.
2. Extremely narrow particle size distribution
Through a single treatment, the D90 of liposomes, nanosuspensions, and other systems can be controlled within 100 nm, and the PDI (polydispersity index) is much better than traditional homogenizers.
3. High repeatability and scalability
Due to the fixed structure of the interaction cavity, laboratory trials (such as 15 mL/min) use the same cavity type as industrial production, with only an increase in the number of parallel connections to achieve "seamless amplification" and avoid the risk of process transfer.
4. Gently handle thermosensitive substances
Although the pressure is high, the action time is extremely short (in milliseconds), coupled with an external cooling system, the temperature rise is controllable, suitable for proteins mRNA、 Sensitive systems such as live cells.
5. No metal pollution
The contact components of the flow path are made of ceramics, tungsten carbide or special alloys, which comply with FDA, USP Class VI and GMP requirements.
3、 Selection and usage suggestions
When choosing a micro jet homogenizer, attention should be paid to:
Work pressure (whether the target particle size requirement is met);
Interaction chamber type and material (Y-shaped suitable for emulsification, Z-shaped suitable for fragmentation; Ceramic corrosion resistance and long lifespan of diamond;
Processing flux and cooling capacity;
Does it support CIP/SIP (online cleaning/sterilization, suitable for GMP workshops);
Attention should be paid during use:
The material needs to be pre filtered to prevent blockage of microchannels;
Control the feed concentration to avoid high viscosity affecting the homogenization effect;
Regularly check the wear of the cavity and replace it in a timely manner to ensure performance.
The microjet homogenizer is not only a homogenization device, but also a "nano bridge" connecting molecular design and product performance. It takes micrometer channels as the stage, completing precise carving of material structures in milliseconds, allowing every nano carrier to carry the hope of precision medicine, and every drop of functional formulation to contain the beauty of technology. In today's rapidly developing global biopharmaceutical and new materials industries, microjet homogenization technology is shifting from optional to standard essential. In the future, with the deepening of personalized medicine and intelligent manufacturing, this precision equipment that operates silently will continue to release transformative power in the journey of human exploration of the microcosm.
