Application plan of single person purification workbench in aseptic inspection and inoculation

The single person purification workbench forms a local sterile environment through an efficient air filtration system, and is widely used for sterile operations in microbial detection, cell culture, and vaccine production. Develop an application plan from three aspects: equipment selection, operating standards, and risk control:
1、 Equipment selection and preparation
Type matching
Choose a vertical or horizontal flow workbench according to operational requirements:
Vertical flow: airflow from top to bottom, suitable for high-risk operations (such as pathogen inoculation), can effectively prevent cross contamination.
Horizontal flow: The airflow is parallel to the operating surface, suitable for experiments that are sensitive to airflow (such as tissue culture).
The equipment must comply with the ISO14644-1 standard and achieve Class 5 (Class 100) cleanliness.
Pre run validation
Before use, turn on the UV lamp to sterilize for 30 minutes, then start the fan to run for 15 minutes. Verify that the suspended particles in the work area are ≤ 3.5 particles/L (≥ 0.5 μ m) through a particle counter to ensure that the environment meets the standard.
2、 Aseptic operating procedures
Personnel and item management
Operators need to wear sterile clothing, gloves, and masks, and wipe their hands with 75% ethanol for disinfection.
After high-pressure steam sterilization, experimental equipment (such as culture dishes and inoculation rings) enter the workbench through the transfer window to avoid direct contamination.
Optimization of operation process
Aseptic inspection: Place the sample to be inspected in the center of the workbench, use sterile forceps to take samples, and avoid touching the inner wall of the workbench.
Inoculation operation: After flame sterilization of the inoculation ring, cool it down and quickly complete the marking or coating to reduce exposure time. Keep items at least 10cm away from the front window during operation to prevent airflow disturbance.
3、 Risk control and maintenance
Pollution prevention and control
Regularly replace the initial and high-efficiency filters (it is recommended to check the pressure difference every 6 months and replace them when it exceeds 20% of the initial value).
After operation, wipe the inner wall of the workbench with 75% ethanol, and seal the waste in a dedicated container.
emergency response
If splashing occurs, immediately stop the operation, cover the contaminated area with disinfectant, clean it up after 10 minutes, and re monitor the environment.
Regular verification
Entrust a third-party testing agency to verify the airflow velocity (0.3-0.5m/s), noise (≤ 65dB), and illumination (≥ 300lx) every quarter to ensure stable equipment performance.
