Microbial limit testing is an important part of quality control in industries such as food, pharmaceuticals, and cosmetics to ensure product safety and quality stability. However, microbial limit testers may sometimes produce abnormal results during the testing process, which not only affects production efficiency but may also mislead product quality assessment. This article will delve into the common interference factors that cause abnormal device detection results and provide detailed troubleshooting methods.
1、 Common interference factors
(1) Characteristics of the sample itself
1. Complex composition: Some samples contain multiple complex chemical components, such as high concentrations of sugars, proteins, oils, etc. These components may inhibit the growth of microorganisms during the detection process or react with the culture medium, affecting the cultivation and counting of microorganisms.
2. Particulate matter: If the sample contains a large amount of insoluble particles, it may clog the filter membrane, hinder the normal interception and transfer of microorganisms, and thus affect the accuracy of the detection results. For example, traditional Chinese medicine extracts containing a large amount of suspended particles are prone to problems during filtration.
(2) Equipment issues
1. Instrument cleaning:Microbial limit testerIf not thoroughly cleaned after use, residual microorganisms, impurities, etc. will accumulate inside the instrument. During the next test, these residual substances may mix into the new sample, resulting in false positive or false negative results.

(1) Processing of samples
For samples with complex components, appropriate pretreatment can be carried out. If diluted to reduce its concentration, most of the particulate matter can be removed by centrifugation or other methods before filtration detection. At the same time, select appropriate filter membrane materials and pore sizes based on the characteristics of the sample to improve filtration efficiency and microbial recovery rate.
(2) Check the instruments and equipment
Regularly conduct comprehensive cleaning and maintenance of the microbial limit meter, disassemble various components according to the manufacturer's instructions, and clean and disinfect them. Focus on checking whether there is any damage or blockage in the filter cup, pipeline, valve and other parts, and replace the problematic components in a timely manner. In addition, regularly calibrate the various parameters of the instrument to ensure that it is in optimal working condition.
(3) Standardize operational procedures
Strengthen the training of operators to familiarize them with and strictly adhere to sterile operating procedures. Prepare the necessary sterile equipment and materials before each experiment, and complete all key operational steps in the ultra clean workbench. For the selection and use of culture media, the appropriate type should be determined based on standard methods and experience, and strictly formulated according to the formula accurately.
