Fluorescence imaging cameras are precision optical instruments, and their performance directly affects the imaging quality and accuracy of experimental data. Reasonable maintenance can not only extend the service life of equipment, but also reduce image deviation caused by pollution, wear and tear, or environmental interference. The following specific maintenance methods are discussed from four aspects: core component maintenance, environmental control, operating standards, and fault prevention:
1、 Deep cleaning of optical system
Lens cleaning: First, use an air blower or soft bristled brush to remove floating dust, and then gently wipe the lens with fiber free lens paper in a circular motion. If encountering stubborn stains, you can dip a small amount of isopropanol (purity ≥ 99%) or specialized lens cleaning solution for treatment. It is strictly prohibited to directly spray the liquid. Prohibit the use of ordinary cleaning agents containing alcohol or ammonia water to prevent corrosion of anti reflective or waterproof membranes. For water stains or mold spots, it is necessary to contact professional personnel to disassemble and handle them.
Filter maintenance: Regularly check whether the excitation light source filter is offset or contaminated. When cleaning, gloves should be worn to avoid finger contact with the surface of the lens and prevent oil residue from affecting the transmittance.
2、 Precise control of environmental factors
Temperature and humidity management: The optimal working environment is a temperature of 1825 ℃ and a humidity of 40% to 60% RH. It is recommended to equip with a constant temperature and humidity box or dehumidifier, especially to avoid condensation water seeping into the lens chamber and causing mold growth.
Light protection: It is strictly prohibited to place the equipment in direct sunlight, as strong ultraviolet rays can accelerate the aging of the plastic casing and cause thermal deformation of internal components. You can install a light shield or choose a black matte bracket to fix the camera.
Electromagnetic interference shielding: Keep away from strong electromagnetic sources such as high-voltage power sources and centrifuges. If necessary, use a metal shielding cover to wrap the signal transmission line. Poor grounding may lead to increased image noise.
3、 Standardized operating procedures
Startup preheating specification: The startup sequence should be to first connect the cooling circulation system before supplying power to the host, and then reverse the operation when shutting down. Ensure sufficient heat dissipation of the laser to avoid high temperature damage to the fluorescent dye.
Focusing protection mechanism: Set limit parameters when using an electric stage to prevent the objective lens from colliding with the sample cell. Manual focusing requires slowly rotating the fine adjustment knob until a slight clicking sound is heard before stopping.
Data backup strategy: Export the original image files to a separate hard drive in a timely manner after each experiment, delete local redundant data to free up cache space. It is recommended to adopt a dual backup mechanism to prevent data loss.
4、 Long term idle disposal plan
Short term shutdown (<7 days): Disconnect the power adapter, cover the body with a dust cover, and seal the front end of the lens with a rubber cover. The storage environment should meet moisture-proof and light proof conditions.
Long term storage (>30 days): In addition to performing the above steps, the battery needs to be separately stored in a dryer and powered on once every quarter to maintain capacitor activity. It is recommended to remotely wake up the device every month to check the sensor response status.
A scientific and systematic maintenance system can keep fluorescent imaging cameras in optimal working condition for 8-10 years, significantly reducing maintenance costs. The key is to establish a standardized operation log system to record every cleaning, calibration, and abnormal phenomenon, providing traceability basis for subsequent maintenance.
