solid-phase extractionSolid Phase Extraction (SPE) is a widely used and popular sample pretreatment technique that uses solid adsorbents to adsorb target compounds from liquid samples, separate them from the sample matrix and interfering compounds, and then use eluent or heating desorption to achieve the goal of separating and enriching target compounds. It is developed on the basis of traditional liquid-liquid extraction using the principle of similar phase solubility based on similar interactions between substances, combined with the basic knowledge of liquid chromatography and gas chromatography stationary phases widely used at present.
Usage guide:
1、 Composition and installation of the device
Core components: including glass vacuum chamber, 12 position SPE panel (including flow control valve), stainless steel drainage needle, vacuum gauge, and sample collection rack.
Installation steps:
Check the integrity of the components and select the collection rack partition as needed.
Install the drainage needle at the bottom of the panel, insert the flow control valve, and secure the support rod.
Connect the vacuum pump to the vacuum flask to ensure sealing (recommended vacuum degree is 254-381mmHg).
2、 Operation process
Sample pretreatment: Filter or centrifuge to remove particulate matter, adjust pH/ionic strength to optimize adsorption.
Activated extraction column:
Reverse phase column: 3-5 mL methanol → 3-5 mL water/buffer, flow rate 1-2 mL/min.
Positive phase column: 3-5mL non-polar solvent (such as n-hexane).
Sample loading and rinsing:
Slowly load the sample (0.5-1mL/min) to avoid overload.
Rinse 3-5 mL with weak elution solvent (such as water or low concentration methanol) to remove interfering substances.
Elution and Collection:
Wash 2-3 times with a strong solvent (such as methanol), with a flow rate of 0.5-1mL/min and 1-2 mL each time.
Collect eluent and concentrate or analyze directly.
