What gases are used in anaerobic incubators

Anaerobic incubators require specific gas mixtures to maintain an anaerobic environment, with common gas ratios as follows:
Applicable scenarios for gas ratio
(1) 90% nitrogen+5% carbon dioxide+5% hydrogen is suitable for strict anaerobic conditions and is suitable for cultivating bacteria that are highly sensitive to oxygen
(2) 80% nitrogen, 10% hydrogen, and 10% carbon dioxide are considered gas combinations for the growth of anaerobic bacteria, which can provide growth conditions for anaerobic bacteria
(3) 95% nitrogen+5% carbon dioxide is suitable for general bacterial and fungal cultivation and is a common gas composition
The function of gas
(4) Nitrogen (N ₂): mainly used to maintain an inert environment inside the box and prevent oxygen from entering.
(5) Carbon dioxide (CO ₂): Maintaining an appropriate acid-base environment is beneficial for the growth of microorganisms.
(6) Hydrogen (H ₂): reacts with residual oxygen to generate water (via palladium catalyst), removes trace amounts of oxygen, and maintains an anaerobic environment.
Precautions
(1) Gas purity: In order to ensure the stability and reliability of the experiment, the gas used should be of high purity.
(2) Safety operating procedures: During use, corresponding safety operating procedures should be strictly followed, such as gradually carrying out ventilation and exhaust to avoid severe reactions causing anaerobic environment imbalance.
(3) Anaerobic catalyst: In the process of gas preparation and use, anaerobic catalysts are required to maintain anaerobic conditions and ensure the accuracy and reliability of experiments.
