The replacement frequency of ion chromatography column protection column is usually 50-200 samples per analysis, or when the column pressure increases significantly. If the sample matrix is complex (such as biological samples), it is recommended to shorten it to 50-100 needles for replacement. Regular replacement can effectively extend the lifespan of the analytical column and ensure the accuracy of the results.
The key to extending the service life of ion chromatography column protection column lies in standardized operation and regular maintenance:
1、 Preparation before use
New column activation: Inject 20-30 times the column volume of eluent (such as carbonate buffer) at a low flow rate of 0.5-1mL/min to remove residual impurities.
System check: Ensure that the pH of the mobile phase is within the column tolerance range (usually 5-9), and the sample needs to be filtered through a 0.22 μ m filter membrane.
2、 Maintenance during operation
Flow rate control: Maintain a regular flow rate of 0.8-1.2mL/min to avoid collapse of the column bed caused by high-pressure impact.
Pressure monitoring: If the pressure suddenly increases by more than 30%, immediately check if the sieve plate is blocked, and use isopropanol reverse flushing (≤ 0.2mL/min).
Sample pretreatment: Complex matrix samples require C18 solid-phase extraction purification to prevent pollutant deposition.
3、 Save after deactivation
Short term discontinuation: Rinse with low flow detergent for 10 minutes daily to prevent microbial growth.
Long term disuse: After emptying, rinse with 20% organic solvent (such as methanol) storage solution and store in a sealed container.
