As the core equipment of life science laboratories, the stability of the full wavelength enzyme-linked immunosorbent assay (ELISA) directly affects the reliability of experimental data. Scientific maintenance can extend the lifespan of instruments and reduce failure rates. The following is a systematic daily maintenance plan:
1、 Daily maintenance: preventive cleaning and basic inspection
1. External cleaning
-Frequency: Execute daily after the end of the experiment.
-Method: Use a dust-free cloth dipped in 75% ethanol solution to wipe the shell, screen, and operation panel to prevent liquid from seeping into the gaps. The display screen needs to be lightly wiped with specialized lens paper to prevent scratches.
-Key areas: Sample compartment door tracks, button gaps, and other areas prone to dust accumulation should be cleaned with a soft bristled brush.
2. Optical system maintenance
-Light source protection: Before starting up, confirm that the cabin door is closed to avoid direct sunlight damaging the deuterium/tungsten lamps. Turn off the light source promptly after the experiment to reduce filament aging.
-Colorimetric dish cleaning (if applicable): Rinse immediately with ultrapure water after use. Stubborn residues can be soaked in neutral detergent. Do not use brushes made of frosted materials. Regularly check for scratches on the transparent surface and replace if necessary.
3. Disinfection of sample area
-Microplate treatment: After the experiment, remove the plate holder, wipe it with deionized water containing 0.1% Tween-20, and then irradiate it with ultraviolet light for 15 minutes
-Emergency spillage: If the sample liquid leaks, it should be immediately dried with absorbent paper and wiped with a special disinfectant to avoid corroding metal parts
2、 Periodic calibration and performance verification
1. Wavelength calibration
-Cycle: Monthly or after replacing the light source.
-Operation: Use standard filters (such as holmium glass filters) to detect the deviation between the actual wavelength and the displayed value, and adjust the grating through software calibration or contacting the manufacturer.
2. Linear validation of absorbance
-Method: Neutral density gradient filter (OD 0.0-2.0) was used to detect linear correlation at various wavelengths (R ² should be>0.999). When deviating, the optical path needs to be recalibrated or the detector needs to be replaced.
3. Cross contamination detection
-Frequency: Conducted quarterly.
-Test: Repeat the detection of absorbance coefficient of variation (CV value) in blank holes, with a requirement of<0.5%. Otherwise, check the sealing of the liquid path or clean the pipette arm.
3、 Environmental Control and Usage Standards
1. Temperature and humidity management
-Ideal conditions: temperature of 20-25 ℃ (± 2 ℃), humidity of 40% -60%. Equipped with independent air conditioning and dehumidifier to prevent condensation water from damaging the circuit.
-Dust prevention measures: The laboratory must achieve ISO level 8 cleanliness and use dust covers to cover the instruments when shutting down.
2. Stable power supply
-Using uninterruptible power supply (UPS), the voltage fluctuation range should be controlled within ± 10%. Do not connect network cables during thunderstorms to prevent induced current surges.
3. Operation taboos
-Violent opening and closing of the sample chamber is strictly prohibited to avoid mechanical misalignment; It is prohibited to directly touch the optical lens, and fingerprints need to be processed with lens cleaning paper.
4、 Long term maintenance and consumables management
1. Replacement of key consumables
-Lamp source lifespan: about 500 hours for deuterium lamps and about 1000 hours for tungsten lamps. Stock up in advance according to usage time records.
-Filter/detector: Detect energy attenuation every two years and replace it when attenuation>20%.
2. Software and data maintenance
-System update: Check firmware versions every quarter and install patches in a timely manner.
-Data backup: Experimental data is backed up to both local servers and the cloud, and audit tracking logs are kept for at least 10 years.
3. Annual deep maintenance
-Performed by manufacturer engineers: Disassemble the optical path module to clean the internal reflector, lubricate the transfer arm guide rail, and test circuit stability.
5、 Fault warning and emergency response
1. Abnormal monitoring
-After daily power on self-test, check the error report and immediately stop troubleshooting if the absorbance drift is greater than 5%.
-When there is an increase in noise or abnormal vibration, check the fan bearings and shock absorbers.
2. Emergency plan
-After a sudden power outage, it is necessary to preheat for 30 minutes to avoid directly restoring unfinished experiments.
