In the development of liquid chromatography methods, some additives are often used to improve chromatographic performance, but these additives are often incompatible with mass spectrometry detection. In some analyses, such as impurity analysis, metabolic analysis, etc., it is often necessary to identify certain newly generated impurities or metabolites by mass spectrometry, while not wanting to change the method selectivity in order to qualitatively identify unknown substances through retention time. However, if the additives in the original method that are not compatible with mass spectrometry conditions are changed, it is likely to cause selective changes, making it impossible to locate the position of the unknown compound to be analyzed.

In response to such issues, Agilent has developed a 2D-LC method based on valve switching, which ensures that the retention behavior of each chromatographic peak does not change (i.e., the original chromatographic conditions remain unchanged), and the mass spectrometry incompatible mobile phase in the original conditions is changed to a compatible mobile phase through two-dimensional liquid chromatography, thereby achieving the purpose of quantitative analysis of unknown impurities using liquid chromatography-mass spectrometry.
