1、 Basic preparation before tuning
Before tuning, it is necessary to ensure that the environmental conditions meet the requirements: the laboratory temperature is controlled at 20-25 ℃, the relative humidity is ≤ 60%, and vibration and electromagnetic interference are avoided. Check the gas system, the purity of helium carrier gas should be ≥ 99.999%, and there should be no leakage in the pipeline; The vacuum system needs to complete at least 10 minutes of vacuuming (high-precision testing is recommended to be extended to 2-3 hours), and subsequent operations can only be carried out after the vacuum degree stabilizes.
2、 Step by step tuning operation process
1. Manual tuning:After starting the software, enter the operation interface and click 'ManualTune' to select the scanning mode (usually scanning the range of m/z 10-100). Focus on observing the intensity of the water peak (H ₂ O) and nitrogen peak (N ₂) - the signal value of the water peak should be maintained at the order of 10 ⁵ (avoiding exceeding E+05), and the intensity of the nitrogen peak should be significantly higher than that of the water peak. When switching to Full mode, ensure that the peak intensity is below 10 ⁵, laying the foundation for subsequent automatic tuning.
2. Automatic tuning:It includes two core steps:
① Leakage inspection: It takes about 4 minutes to determine the sealing of the system by monitoring the ratio of N ₂ and H ₂ O. If the height of the N ₂ peak exceeds twice that of H ₂ O (i.e. the N ₂ peak exceeds the interface window), it is determined that there is air leakage, and the vacuum must be immediately released and the sealing of each interface checked.
② Standard tuning: The process takes about 32 minutes, and the system automatically optimizes parameters after selecting EI mode. After successful tuning, key indicators need to be verified: abundance value>10 ⁷ (such as characteristic peaks of 69, 131, 218, 414, 502, etc. are clearly visible), base peak stability meets the requirements, and the instrument enters the optimal detection state.
3、 Precautions
During the tuning process, it is necessary to strictly avoid starting the ion source without vacuuming, and regularly cleaning the ion box can maintain sensitivity. If the voltage of the MultiplerVoltage drops after cleaning the ion source, it is a normal phenomenon, but if it continues to be lower than the factory value, maintenance is required. Standard tuning must be performed every startup and every 15 days of operation to ensure the reliability of long-term testing data.
