Determination of Tetramethylenediamine Content in Fasudil Hydrochloride Drug by Ion Chromatography

Fasudil hydrochloride, as a vasodilator, can effectively alleviate cerebral vasospasm and is a novel drug with a wide range of pharmacological effects [1-2]. Gao is an intermediate in the synthesis process of hydrochloric acid fasudil [3]. The method reported in literature for determining hydrochloric acid fasudil is liquid chromatography [4], but there are few reports on the determination of the impurity content of tetramethylenediamine. The main reason is that the content of tetramethylenediamine is relatively low, and it is weakly retained on conventional reverse phase chromatography columns. At the same time, there is no UV absorption. Therefore, this article uses ion chromatography with conductivity as the detector to determine the content of tetramethylenediamine in hydrochloric acid fasudil drugs. The results showed that under low concentration conditions, tetramethylenediamine was adsorbed on the pre-treatment column. However, when the concentration was greater than 15 mmol/L, the peak area of tetramethylenediamine was consistent with that of direct injection (see Table 1). Therefore, 17 mmol/L LMSA with the same concentration as the mobile phase was used as the * solution for dissolving the sample
