What are the details to pay attention to when replacing a liquid chromatography column
Date: 2025-10-20Read: 2
The liquid chromatography column is the core separation component of high-performance liquid chromatography (HPLC), and its performance directly affects the accuracy and repeatability of analysis results. When abnormal peak shape, increased column pressure, decreased theoretical tray number, or loss of stationary phase occur, the chromatographic column should be replaced in a timely manner. The following is the standardized replacement process and key precautions:
1、 Preliminary preparation
safety protection
Wear protective gloves, goggles, and lab coats to avoid contact with organic solvents and sharp parts.
Turn off the instrument power and release the system pressure to prevent high-pressure fluid splashing.
Tools and Materials
New chromatographic column (model/size consistent with the original column), two-way connector, wrench, sealing gasket (PEEK material preferred).
Spare pipeline accessories (if length adjustment is required).
Refer to the instrument manual and chromatographic column technical parameter table.
System status confirmation
Stop the current running program, switch to pure mobile phase (such as methanol/acetonitrile), and continuously rinse at low flow rate (0.5-1mL/min) for at least 30 minutes to remove residual samples from the pipeline.
Disconnect the detector signal line and mark the position of the inlet and outlet pipeline interfaces.
2、 Dismantling steps for old columns
Positioning chromatography column module
Find the chromatographic column bracket inside the column temperature chamber and observe the sequence of pipeline connections (usually: injection valve → chromatographic column → detector).
Gradually remove the old pillars
Loosen the nuts at both ends with a wrench, first remove the outlet end (near the detector side), and slowly release the residual liquid into the waste liquid bottle.
Wait for a moment to release the system pressure before disassembling the inlet end.
Do not forcefully pull or tug on the pipeline to prevent damage to the joint threads.
Cleaning interfaces and evaluating wear and tear
Check if the sieve plate is blocked or deformed, and clean it with ultrasonic waves if necessary.
Record the old column serial number, usage duration, and fault characteristics for future selection reference.
3、 Key points for installing new columns
Direction verification
All chromatography columns have clear flow direction markings (arrows pointing to the outlet), and reverse installation can cause the packing to collapse.
Partial reverse phase columns can be used at both ends, but positive phase/ion exchange columns must strictly follow the direction.
Bubble free installation
Adopting the "liquid docking method": immerse the inlet of the new column into a beaker containing the mobile phase, and connect the pipeline while venting.
Start the pump and inject at a very low flow rate (0.2mL/min) until there are no bubbles discharged from the outlet.
Torque control
Use a specialized wrench to alternately tighten the nuts diagonally, with moderate force (a slight resistance to the touch is sufficient). Overtightening may cause compression of the column bed.
It is recommended to regularly check for looseness during operation after installation.
4、 System restart and balancing
Initial flushing
Rinse at low flow rate for 60 minutes with initial mobile phase composition (mixed in the same proportion) and observe column pressure stability.
If the column pressure is abnormally high, pause and rinse briefly with isopropanol to clear the blockage.
Gradient equilibrium
Execute the blank gradient program and monitor the baseline drift to be<± 0.5mAU.
Inject the test mixture to verify the separation degree and compare whether the theoretical number of trays meets the standard (N ≥ 80% of the theoretical value).
data calibration
Update the chromatographic column information in the workstation method file and rebuild the retention time comparison table.
Save the new column certification report, including column efficiency test chart and pressure curve.
Standardized operations can maximize the lifespan of chromatography columns, with a typical reverse phase column lifespan of up to 500-1000 injections. Daily maintenance recommendations include simple weekly flushing and monthly deep regeneration maintenance.
