NanoCoulter evaluation of five extracellular vesicle separation methods: high efficiency and purity comparison
Date: 2024-10-21Read: 1
In recent years, extracellular vesicles (EVs) have attracted much attention due to their important role in biomedical research. They not only play a crucial role in intercellular communication, but also demonstrate enormous potential in early diagnosis and treatment of diseases. However, how to effectively and high-quality separate these vesicles remains a challenge. In a recent study, researchers conducted a comprehensive evaluation of the quality and efficiency of five commonly used extracellular vesicle separation methods using a NanoCoulter particle analyzer based on resistance pulse sensing (RPS) technology[1]This provides valuable reference for researchers. Relevant reports have been interpreted in detail on the official account. What is an extracellular vesicle

图片

EVs refer to nanoscale vesicles released from cells, widely present in bodily fluids such as blood, urine, saliva, etc. These vesicles are rich in functional proteins, small molecules, nucleic acids, and other metabolites. EVs are present in almost all types of cells and body fluids, including plasma, urine, saliva, and breast milk. The types and contents of proteins, lipids, and miRNAs carried by EVs can directly reflect the type of their source. The study of EVs contributes to a better understanding of cellular physiology and pathology, and in clinical practice, EVs not only demonstrate enormous potential in disease diagnosis, but may also become a new tool for drug delivery. Therefore, the separation and characterization methods of high-purity EVs that meet specific experimental requirements have become particularly important.

Comprehensive comparison of five separation methods

图片

In this study, researchers conducted a detailed evaluation of five common EV separation techniques, including:

1.Traditional differential ultracentrifugation (UC) methodA speed above 30000 r/min is called overspeed centrifugation. On the basis of ultracentrifugation, gradually increasing the centrifugation speed can be used to separate samples of different sizes, which can effectively separate high-purity EVs and is often considered the "gold standard" for nanoparticle extraction.

2.Ultrafiltration (UF) methodThe rapid selective separation of samples is achieved through the filtration of microporous structures in semi permeable membranes, which is suitable for large-scale production.
3.ExoEasy membrane affinity methodUsing affinity ligand modified membranes (affinity membranes) as a medium for separation, the operation is simple.
4.QEV column chromatographyBased on size exclusion chromatography, separation is performed according to the molecular size of the tested component, which is suitable for separating EVs with uniform size.
5.ExoQuick polymer precipitation methodThe method of using high molecular weight polymers (such as PEG) to alter the solubility and dispersibility of EVs, causing them to aggregate and precipitate in solution, and then precipitating them by centrifugation. This method has a short separation time, simple operation, and is relatively mild for EVs.

RPS technology

图片

Resistance Pulse Induction (RPS) technology is an emerging technique for characterizing nano single particles and is currently based on electrical principlesParticle characterizationmethod. Evaluate the size and concentration of extracellular vesicles and zeta potential by measuring changes in current through tiny nanopores. This method has the advantages of accuracy, speed, simplicity, and high sensitivity, making it an ideal tool for evaluating the quality of vesicle separation methods.

separationThe secret behind it: the debate between efficiency and purity

图片

Through RPS technology, researchers analyzed in detail the particle size distribution, particle concentration, and purity of each separation method. The results indicate that although high particle concentrations can be obtained using ultrafiltration, qEV column chromatography, and ExoQuick methods, traditional differential ultracentrifugation performs the best in purity.

particle size distributionThe particle size of EVs separated by UC, UF, exoEasy, and qEV is relatively consistent, about 71 nm, while the particle size of EVs separated by ExoQuick is slightly larger, with an average of 77 nm.

Particle concentrationUltrafiltration (UC) showed the highest particle concentration, reaching 2.10 × 10 ^ 312/mL, The particle concentration of UC method is relatively low, at 7.33 × 1010/mL。

purityThe EV purity of UC method is the highest, with a particle count of 7.88 × 10 per milligram of protein11.

How to choose the appropriate separation method?

图片

Each separation method has its own characteristicsadvantage According to research results, although ultracentrifugation is complex and time-consuming, it is still the first choice when higher purity EVs are required for experiments; However, for application scenarios that pursue high efficiency and large-scale production, ultrafiltration and qEV column chromatography provide faster separation speeds and higher particle concentrations.

Understanding the advantages and disadvantages of different separation methods will help researchers better conduct experimental research and application development related to extracellular vesicles. This article comprehensively evaluates five extracellular vesicle separation methods using RPS technology and the precise performance of NanoCoulter, providing scientific basis for researchers to choose suitable separation techniques.

The selection of appropriate separation methods should consider the specific requirements of experimental needs, including separation efficiency, purity, and ease of operation. The same goes for selecting appropriate particle characterization methods,In the above study, NanoCoulter demonstrated the following advantages in characterizing EVs:

图片

Advantage 1: Single particle detection, precise and interference free

NanoCoulter, with its advanced nanoscale RPS technology, achieves precise capture and analysis of every nanoparticle.Imagine that in complex biological fluids, every tiny EVs passing through nanopores cannot escape its' comprehensive examination '.Breaking the phenomenon of large particle signals shielding small particles and resulting in distortion in traditional optical methods.Especially for multi dispersed EVsBiological fluid samples, whether in size, concentration, or zeta potential, each data is derived from direct measurements of individual particles to ensure the authenticity and reliability of the results. This precision can take the study of cell vesicles to a new level!

图片

Advantage 2: High precision, multi parameter analysis

01

NanoCoulter can comprehensively analyze the particle size, particle size distribution, particle concentration, and zeta potential of EVs.Accurate particle size distribution analysis helps us understand the true size distribution of EVs samples, while particle concentration reflects the abundance and separation quality of EVs. Zeta potential is closely related to the stability of EVs and the targeted drug delivery of engineered EVs.These data provide us with comprehensive information on EVs, providing solid data support for nanomedicine scientific research and clinical applications.

图片

Advantage 3: Adapt to complex samples

01

Unlike traditional optical methods that analyze particles in the sample as a whole, NanoCoulter performs well in complex and dispersed samples. It can accurately characterize nanoparticles in low refractive index, high ion concentration, or low fluidity liquids, ensuring reliable results even in complex biological fluids, and is professionally suitable for research in the field of biomedicine.

图片

Advantage 4: Fast and convenient operation process

01

The operation process of NanoCoulter is simple and fast, without the need for complex sample pretreatment, as well as equipment thermal and calibration steps. Each nanopore chip undergoes strict calibration and verification during production, and there is no need to consume standard products at the user end. This enables researchers to obtain results faster and accelerate the pace of scientific discovery.

图片

Advantage 5: Widely applicable

01

The application areas of NanoCoulter are far beyond EV research. It has demonstrated enormous potential and value in multiple fields such as biomedicine, materials science, and environmental monitoring. With its high precision, high stability, and wide applicability, NanoCoulter is gradually becoming a powerful assistant in the hands of researchers.
