Ion chromatography pretreatment columnIt is a key component that ensures the accuracy of analysis results, and its performance directly affects detection sensitivity and data reliability. During use, the following precautions should be taken:
1、 Installation and Connection
Correct installation: The pre-treatment column should be installed in the chromatography column box, ensuring that the sealing ring is placed correctly to prevent liquid leakage. The inlet and outlet should be tightly connected to avoid pressure loss.
Handle with care: Avoid severe collisions or twisting during installation and disassembly to prevent column damage or decreased sealing.
2、 Mobile phase selection and processing
Mobile phase requirements: It is recommended to use filtered ultrapure water (resistivity>18M Ω) as the mobile phase to avoid particle blockage of the column. If organic solvents need to be used, their quality must be ensured and replaced regularly.
PH adjustment: Adjust the pH value of the mobile phase according to the analysis requirements to avoid damage to the column packing caused by extreme pH conditions.
Bubble removal: The mobile phase needs to be removed by ultrasound, filtration and other methods to prevent abnormal peak shape or column pressure fluctuations.
3、 Sample pre-treatment
Impurity removal: Select precipitation, filtration, centrifugation and other methods based on the properties of the sample to remove suspended particles and prevent column blockage.
Representative guarantee: Ensure that the sample processing process does not alter its composition to obtain accurate analytical results.
Dilution and filtration: High concentration samples need to be diluted and filtered to avoid the accumulation of pollutants that may affect the next analysis.
4、 Operation process control
Column pressure monitoring: Real time monitoring of changes in column pressure. If there is a sudden increase, it may be due to blockage of the mobile phase or damage to the chromatographic column. Injection should be stopped immediately and checked.
Temperature control: Maintain a constant temperature operation to avoid temperature fluctuations affecting the viscosity and separation efficiency of the mobile phase.
Injection volume control: Ensure moderate injection volume to avoid overload or insufficiency; The injection needle needs to be cleaned to prevent residue from interfering with the analysis.
Flow rate control: Adjust the flow rate according to the column specifications (such as 2mL/min) to ensure that the sample is fully purified and the packing is not damaged.
Discard initial solution: Discard the first few milliliters of sample that has passed through the column during injection (such as discarding 3mL of 1mL specification column) to avoid the influence of activation solution or impurities on the results.
5、 Safety and Protection
Organic solvent safety: When using organic solvents, keep them away from sources of fire and store them in a cool and ventilated place to prevent the risk of flammability and explosion.
Personal protection: Wear protective equipment such as goggles and laboratory gloves during operation to avoid contact with corrosive or toxic substances.
6、 Maintenance and upkeep
Daily cleaning: After use, rinse the column in reverse with pure water or specified solvent to remove particles or chemical residues. High concentration samples need to be cleaned immediately after detection.
Stubborn pollution treatment: For stubborn pollution, the flushing time can be extended or gradient elution can be used, but strong corrosive reagents should be avoided.
Idle storage:
Short term idle: Rinse with pure water and soak in protective solution to prevent the packing from drying out or breeding microorganisms.
Long term storage: After drying the column, seal and store it in a cool and dark place to avoid high temperature or humidity that accelerates material aging.
Re enable: After being idle for a long time, it is necessary to thoroughly rinse with mobile phase to restore the performance of the column.
7、 Regeneration and Reuse
Regeneration method:
Reverse phase column (such as C18, RP): Wash the organic matter with 20mL of methanol, then rinse with 10mL of water to avoid cross contamination.
Ionic columns (such as H, Na, Ag, Ba): Wash the column with 10mL of water and determine whether it can be reused based on the color change of the packing (such as when the Ag column turns black after adsorbing Cl ⁻ and needs to be replaced when 3/4 of it changes color).
Usage limit: It is recommended that the reverse phase column be reused no more than 3 times, while the ion type column should be used based on the packing capacity and sample impurity content to calculate the usage frequency.
8、 Performance testing and replacement
Regular testing: Determine whether the column efficiency has decreased by comparing the retention time, peak shape, and separation degree of standard samples.
Timely replacement: If a decrease in separation efficiency or abnormal baseline fluctuations are found, the cause should be investigated and a new column should be replaced.
