The role of SPR molecular interactor in drug development process
Date: 2025-08-13Read: 4
SPR molecular interactor is a real-time monitoring technology based on optical principles, widely used in fields such as biology, chemistry, drug development, etc. It has irreplaceable advantages, especially in studying the interactions between molecules. By measuring the interactions between molecules on solid surfaces and molecules in solution, high-sensitivity real-time data can be obtained.

  SPR molecular interactorWorking principle:
1. Stimulate surface plasmon resonance
When a laser beam is irradiated onto a metal surface at a certain angle, if the incident angle meets certain conditions, the light energy will excite free electrons on the metal surface to generate plasma waves, thereby forming surface plasma waves on the metal surface. At this point, some of the light energy is converted into surface plasmon waves, resulting in a decrease in reflected light intensity.
2. Molecular binding and changes in reflected light
In SPR experiments, metal surfaces are typically modified with a layer of biomolecules (such as antibodies, receptors, etc.) that can specifically bind to target molecules in solution. When the target molecule binds to biomolecules on the metal surface, it changes the refractive index near the surface, thereby affecting the resonance angle of the surface plasmon. By monitoring the changes in reflected light intensity in real-time, dynamic information on molecular interactions can be obtained.
3. Data analysis
SPR instruments monitor real-time changes in reflected light intensity by altering the angle of incident light. When molecules bind to metal surfaces, the refractive index changes, resulting in a change in the intensity of reflected light. By analyzing the relationship between reflected light intensity and incident angle, important parameters such as the rate constant of molecular binding (kon) and dissociation rate constant (koff) can be obtained to evaluate the strength and affinity of intermolecular interactions.
Application areas of SPR molecular interactor:
1. Research on the Interaction of Biomolecules
Capable of real-time and label free detection of interactions between biomolecules. Researchers can monitor the affinity and kinetic parameters between antibodies and antigens, enzymes and substrates, receptors and ligands, and other molecules through SPR technology. For example, in drug screening, SPR technology can be used to study the binding characteristics of small molecule drugs with target proteins.
2. Drug discovery and development
In the process of drug development, it can help researchers evaluate the binding affinity between candidate drug molecules and target proteins, and then screen potential drug molecules. In addition, SPR can also be used to monitor the metabolic process of drugs and study the dynamic changes of drugs in the body.
3. Immunological research
SPR technology can be used to study the binding and dissociation processes between immune molecules (such as antibodies and antigens), providing important data for vaccine development and immune detection. Through SPR technology, researchers can quantify the affinity of immune molecules and evaluate the strength of immune responses.
4. Environmental monitoring and food safety
It can also be used to detect harmful substances in environmental factors such as water quality and air quality. Through specific biosensors, SPR technology can quickly and accurately detect pollutants. In addition, SPR technology can also be used for food safety testing, such as detecting pathogenic bacteria or toxic substances in food.
5. Clinical testing
In clinical medicine, it can be used for the detection of biomarkers, such as tumor marker analysis in early cancer diagnosis. The high sensitivity of SPR technology enables it to play an important role in detecting biomolecules at low concentrations.
