As a commonly used analytical instrument in laboratories, the accuracy and precision of ultraviolet spectrophotometers are affected by various factors, mainly including the instrument's own performance, operating process, and environmental conditions.
Instrument performance is the fundamental guarantee. The stability of the light source is crucial. If the intensity of the light source fluctuates, it can lead to unstable absorbance measurements and affect accuracy. For example, deuterium lamps and tungsten lamps are commonly used light sources, and their service life and luminescence stability directly affect the performance of the instrument. If they are not replaced in a timely manner after a period of use, the measurement results may deviate. The sensitivity and linear range of the detector cannot be ignored. Insufficient sensitivity makes it difficult to detect low concentration samples, and exceeding the linear range can distort the measured values.
The operation process has a significant impact on the results. Improper sample processing can introduce errors, such as uneven sample and particle impurities, which can increase light scattering and result in higher absorbance measurements. The selection and use of colorimetric dishes are also crucial. Different materials of colorimetric dishes are suitable for different wavelength ranges, and their transparency varies. Improper selection or unclean or scratched colorimetric dishes can interfere with measurements. In addition, failure to regularly perform wavelength calibration and absorbance calibration during the instrument calibration process can cause measurement results to deviate from the true values.
Environmental conditions cannot be ignored either. Temperature changes can affect the electronic component performance of the instrument and the refractive index of the solution, thereby affecting the measurement results. Excessive humidity may cause optical components to become damp and moldy, reducing light transmittance. Vibration and direct sunlight can also interfere with the normal operation of the instrument, making the measurement data unstable.
To ensure the accuracy and precision of the UV spectrophotometer, regular maintenance of the instrument, standardized operating procedures, and strict control of experimental environmental conditions are necessary to obtain reliable analysis results.
